What would the ability to simultaneously quantitate 50 cytokines or other soluble
biomarkers from a single 25 μL sample do for your research? The use of multiplex
immunoassays for multiple analyte detection has proven to be an invaluable tool for the
comprehensive study of biological systems. As these systems are comprised of networks
of secreted proteins, including cytokines, chemokines, growth factors and other proteins,
multiplex assays are an efficient method for biomarker profiling of a large set of proteins
from a small sample.
ProcartaPlex Multiplex Immunoassays use the Luminex® xMAP (multi-analyte profiling)
technology that enables the simultaneous detection and quantitation of up to 50 protein
targets in a single 25-50μL sample. The technology employs the use of differentially dyed
capture beads for each target to allow for a multiplex “ELISA-like”assay. ProcartaPlex
allows you to profile up to 50 times more analytes using significantly less sample in the
same time that it takes to perform an ELISA. Refer to pages 2-3 for a technology overview
and assay workflow principles.
ProcartaPlex Multiplex Immunoassays are available in multiple formats across six species (Human,
Mouse, Rat, Non-Human Primate, Porcine and Canine) to meet the needs of your research.
ProcartaPlex Panels—Pre-configured biologically-defined and disease-defined

panels using magnetic beads for the quantitative multiplex analysis of 6 to 45 analytes
in a single sample of plasma, serum, cell culture supernatant, or other bodily fluids.
ProcartaPlex Simplex—Magnetic bead sets to detect individual analytes that are

designed to be added to ProcartaPlex panels to increase customization. Alternatively,
multiple ProcartaPlex Simplex sets can be combined and run using the ProcartaPlex
Basic Kit which includes all non-target specific reagents needed to perform the
ProcartaPlex assay.
Procarta® Mix&Match Assays—Custom blended and optimized panels to deliver

results that are tailored to the panel design of YOUR choice and your sample type.
Simply select the desired analytes from a Procarta Mix&Match list, then select sample
type, bead type, and instrument type. A custom assay kit will be built and optimized
for your requirements.

Technology Overview
xMAP® Technology
The xMAP technology utilizes two bead types—magnetic and polystyrene. The magnetic
MagplexTM bead is a superparamagnetic 6.5 micron microsphere with a magnetic core and
polystyrene surface. The polystyrene MicroplexTM bead is a 5.6 micron polystyrene microsphere.
Both bead types are internally dyed with precise proportions of red and infrared fluorophores.
The differing proportions of the red and infrared fluorophores result in 100 unique spectral
signature microspheres which are identified by the Luminex® xMAP detection systems. The
conjugation of a distinct monoclonal antibody to a distinct bead allows for analysis of multiple
analytes in a single well.
The polystyrene bead requires expensive, and sometimes difficult to use filter plates. This bead
type also makes automating the assay more difficult. The magnetic bead provides significant
advantages over the polystyrene bead because it allows for greater ease and flexibility. This
ease is realized in the ability to use a flat-bottom plate with either a hand-held magnetic plate
washer (cat. no. EPX-55555-000) or an automated magnetic plate washer. In addition, a filter
plate/vacuum manifold system, commonly used with polystyrene beads, can also be used with
magnetic beads.

Luminex® Instrumentation
Luminex instrumentation is comprised of three instrument models—MAGPIX, LX200 and
FlexMAP 3D. These instruments are based on either the principles of quantitative fluorescent
microscopy or fluorescent flow cytometry. The multiplex quantitation capability of an
assay is dependent upon instrument model, and ranges from 50 analytes with use of the
MAGPIX, to 500 analytes with the FlexMAP 3D. Used in combination with ProcartaPlex, the
Luminex platform delivers fast and cost-effective immunoassay profiling analysis.

Th1/Th2 T Helper Cell Cytokine Panels
Th1 and Th2 lymphocytes are critical mediators of cellular and humoral (antibodymediated) immune responses, respectively. Th1 cells play an important role in host
defense systems for intracellular microbial agents and viruses. Th1 cell promoting factors
include IFNγ and IL-12(p70) and are the primary source for the inflammatory cytokines
lFNγ, IL-2, and TNFβ (LTα) that stimulate other defense cells for the destruction of bacterial
pathogens. These cytokines also help foster the development of cytotoxic lymphocytes
that are responsible for the cell-mediated immune response against viruses and tumor
cells. Alternatively, Th2 cells mediate the humoral response by producing various cytokines
such as IL-4, IL-5, IL-6, IL-9, IL-13 and IL-17E (IL-25). Some of these cytokines counteract the
Th1 responses. Functionally, Th2 cells stimulate and recruit specialized subsets of immune

cells to the site of infection or in response to allergens or toxins.
Due to the central role of Th1 cells in the immune system, over activation or misdirected
activation also makes them key players in Th1-dominant autoimmune diseases such as multiple
sclerosis, type-1 diabetes, rheumatoid arthritis and delayed-type hypersensitivity responses.

Tfh/Th9/Th17/Th22/TReg T Helper Cell
Cytokine Panels
In addition to the Th1/Th2 cytokine expression profiles, there are many other CD4+ T Helper
cell types that also have distinctive differentiation and secretion cytokine profiles. Each of
these different cell types have very important roles in the body’s immune response. Th17 and
TReg cell types have gained the greatest amount of interest over the last decade with Tfh now
gaining considerable interest. Other T helper cells, such as Th9 and Th22, also exist.
Th17 cells act as a bridge between adaptive and innate immunity. Cell differentiation
is driven by TGFβ and IL-6 in vitro, with IL-1β and IL-23 also being necessary in vivo for
Th17 development. Th17 cells require IL-23 for expansion and stabilization while the
cytokines IFNγ, IL-27 and IL-4 inhibit differentiation. Activated Th17 cells produce the
cytokines IL-17A, IL-17F, IL-17AF, IL-21, IL-22, IL-26 (human), GM-CSF, MIP-3α and TNFα.
Functionally, Th17 cells play a key role in host defense against extracellular microbes and in
autoimmune disease. Its inflammatory responses have been implicated in a broad array of
inflammatory and autoimmune diseases.
Regulatory T (TReg) cells function to maintain self-tolerance and immune homeostasis
by suppressing the activation, proliferation and effector functions of various immune
cells. One mechanism for this involves the secretion of IL-10 which inhibits effector T cell
responses. TReg cells also secrete IL-35 and TGFβ that induces conventional CD4+ T cells
to differentiate into TReg cells, thereby skewing the ratio of TReg to T helper cells during
an immune response. Understanding the mechanisms by which TReg cells exert their
suppressive function has broad implications for drug development strategies aimed at
treating cancer, diabetes and other autoimmune diseases.
T follicular helper (Tfh) cells are essential in the development of antigen-specific
effector and memory B cell responses. They regulate humoral immunity and play a role
in the development of long-term antibody responses. They require IL-6, IL-21 and B cell
interaction for complete development. Upon antigen-specific stimulation, Tfh cells release
IL-4, IL-10, IFNγ and IL-21 to stimulate mature B cells into forming germinal centers and
undergoing antibody class-switching.
Th9 cells are characterized by the secretion of IL-9. Th9 cells differentiate with the
combination of TGFβ and IL-4, while the cytokines IL-1β, IL-6, IL-10, IL-21, IFNα and
IFNβ enhance IL-9 expression of cultured Th9 cells. Conversely, IFNγ and IL-27 inhibit its
production in human Th9 cells making it possible that Th1 response may suppress Th9 cell
differentiation. Th9 cells are also known to be capable of inducing tissue inflammation,
Experimental Allergic Encephalomyelitis (EAE), and play a role in allergic asthma.
Th22 cells are a subset dedicated to the production of IL-22, an IL-10 family member.
Maturation of Th22 cells requires TNFα and IL-6. Th22 cells are characterized by the
expression of IL-22, IL-13 and other factors including FGF isoforms involved in tissue
remodeling. Th22 cells appear to provide a protective role in regulating wound repair and
healing in the skin, gut and lungs.
