The word “aging” encompasses a range of
physiological changes in humans, human tissues,
and constituent cells. Alterations in metabolism,
whether at the level of small molecules, protein
homeostasis, signaling patterns, or intertissue
communication, underpin many of the processes
that ultimately decline or shift during aging. The
recent wealth of information on how perturbation
of metabolic factors can accelerate or impair aging
processes makes this an exciting time to convene
and consider the new biology being uncovered,
its potential impact on human health, and the
implications for the coming age of precision
medicine.

Phagocytes—of which the macrophage is the
prototypical cell but which also include a variety of
other cell types such as microglia, dendritic cells,
fibroblasts, and osteoclasts—play a broad role
in host homeostasis in addition to host defense.
Their roles include underappreciated functions in
development and aging, tissue remodeling during
morphogenesis, repair, and malignancy.
In 2006, Shinya Yamanaka and Kazutoshi
Takahashi reported the Nobel Prize winning
discovery of induced pluripotent stem cells
(iPSCs) in Cell. In the 10 years that have passed
since, iPSC technology has provided fundamental
insights into our understanding of cell fate,
mammalian development, and human disease.
It has also spurred renewed interest in direct
reprogramming across lineages, and this area too
has flourished. Progress in understanding the
biology of iPSCs and reprogramming has moved
in parallel with work on clinical translation to apply
the insights obtained, and the first iPSC-based
clinical trials are already underway.
2016 marks the centenary of the death of
Elie Metchnikoff, the Russian scientist who
first discovered phagocytes. This centenary
coincides with a surge of new molecular and
cellular information about phagocytes that
crosses traditional disciplinary boundaries. This
renewed interest in phagocytes provides an ideal
starting point for a meeting of cell biologists,
immunologists, and developmental biologists
interested in further exploring the phagocyte’s
fundamental role in tissue biology, identifying
gaps in our current knowledge, and defining
disease pathways, beginning with infection and
inflammation and extending beyond. The projected
program will provide an in-depth analysis of a wide
range of topics not covered in any meeting hitherto
or planned for the coming years.

Throughout most of the latter half of the 20
century, research on RNAs focused predominantly
on their key role as intermediaries between
DNA and protein. It was Jacob and Monod who
originally proposed that RNAs, because of their
intrinsic specificity, might be the elusive factors
that instruct genes to turn on and off. In the end,
these regulatory factors turned out to be proteins,
now commonly referred to as transcription factors.
Yet, with the discovery of small RNAs in the 1990s,
a wave of new research on RNAs has resurrected
the idea that RNAs can also serve regulatory
functions.
