In a series of experiments , it was found that
exposure of ‘cut’ hair to cocaine vapor and to
aqueous solutions of cocaine HCl resulted in
significant contamination of hair samples.

WASH: After adding 1 ml of methanol wash
solvent, the hair was cut into fine particles with
surgical scissors. The total time of cutting and
washing lasted approximately 1 min…..The
methanol wash fraction was collected and the
remaining hair was rinsed with methanol ( 2 X 1
ml) under vacuum. The wash fractions were
combined….

A Comparison Study of Two Wash Procedures
Using Cocaine Contaminated Human Hair. Wang et al 1999
14 drug free control human hair samples were soaked in
a 1000 ng/ml cocaine solution.
2 different wash procedures, a NIDA methanol wash and
the PMD wash procedure were compared.
All washes and hair digests were analyzed by LC/MS/MS
The NIDA methanol wash procedure removed 49.8%,
49 8%
while the PMD wash procedure removed 88.4% of the
externally contaminated cocaine.
An extended wash p
procedure removed about 3% more.
A comparison of the data showed that the PMD wash
procedure was able to differentiate between external
contamination, but the methanol wash procedure
showed 8 false positive findings.
Careful washing of the hair samples is mandatory in any
laboratory setting where human hair is being tested.
