Valley Agricultural Software 559-686-9496 | [email protected] | www.vas.com 4 2014 ANNUAL REPORT DEDICATED TO GARY MURPHY The 2014 Tulare Dairy Herd Improvement Association’s Annual report is dedicated to longtime employee Gary Murphy for his 43 years of service as a tester for the Association. Gary was living in the Chino area when he was drafted to the military in 1965 and served in Korea during 1966. After the service it was the in Sun Valley, Ca where Gary was first introduced to cow testing through his father-in-law and went to work for George Andrews Testing Lab. Four years later he learned of an outfit called DHIA. Gary applied for a testing job at the once named Tulare County Dairy Herd Improvement Association in May of 1972 with experience using Milk-O-Meters and testing cows. Manager Charles Latham hired Gary and took him to Taggart Farms to see what he knew about testing. After 2 hours in the barn, Mr. Latham left Gary to complete the job on his own and the rest is history. Murphy is an extremely dedicated employee and can work with anyone, never turns down work and truly enjoys his job. He treats management, dairymen, milkers and coworkers with respect. Besides his family and work Gary has two other passions, bowling and bass fishing. Gary bowls in a league and carries a 203 average! He is also a member of the 10 pound club. On behalf of the Tulare Dairy Herd Improvement Association, it is with great honor that the 2014 Annual Report be dedicated to Gary Murphy for his 43 years of continued dedicated service as a tester for the association and the dairymen of Tulare DHIA. 5 Our team of experts know the dairy industry like no other accounting firm and have been helping businesses like yours for almost 100 years. Call us today to discover a fresh approach. 6 PRESIDENT’S MESSAGE Ed Brower At the beginning of a new year it is always wise to look back and reflect on the year’s events. We can do so this year, with thanks as it was a fantastic time to be in the dairy industry. Milk prices were at an all-time high and feed costs lower than previous years. A time to conserve and prepare for what may lie ahead. Do we know what lies ahead? No! But, we’re facing severe drought conditions and once again milk prices which are decreasing. Remember to stand firm together as we fight the battle for water use in agriculture production. Tulare DHIA continues in over 100 plus years of service, providing the tools necessary to equip dairymen to do their best. In 2014, the board approved the purchase of a new Bentley sampling machine which will increase our efficiency and update some of our aging machinery. While this was a major expense and the cost of doing business continues to rise, we did not increase sampling costs for our members this year. Dave and Tony are continuing to lead the association and manage with the best business practices. Our supervisors, lab personnel, and employees continue to be dedicated to their work, are excellent at taking accurate samples and reporting the results in a timely fashion in order that you may make the best business decisions for your milking herds. Congratulations to the award winners and scholarship recipients on their hard efforts. Also, thank you fellow board members for taking time out of your busy lives to serve in this association. I would encourage those who have an interest in serving to contact a board member. Finally, I ask the Lord to continue to bless us all this year and beyond. She has also been in Link Crew Leadership, the Tulare Western ASB and was Junior Class President. Hannah also took 1st place on the written Dairy Knowledge Exam and a member of the 2nd place California Junior Dairy Bowl Team and 3rd place in Prepared Public Speaking at the 2013 National Holstein Convention. She was also a member of the 2nd place California Senior Dairy Bowl Team at the recent 2014 National Holstein Convention. The Anti-Mullerian hormone is a glycoprotein first described to have an important role in sex differentiation in early fetal life. It is produced by Sertoli cells in the male gonad and by granulosa cells mainly from early antral follicles during the growing phase of follicular waves in the female ovary (Monniaux et al., 2014). Thus, because number of antral follicles seem to be variable but fairly repeatable within cattle (Ireland et al., 2007) and because AMH is a hormone produced by granulosa cells of growing follicles – which are correlated with the entire population of follicles in the ovary, AMH has been used to indirectly measure the size of the follicular population in cattle (Monniaux et al., 2014). Another advantage when utilizing AMH to predict follicular pool is that AMH levels are fairly constant throughout the estrous cycle and at different physiological states of the animal (Souza et al., 2014 – in press; Rozner and Verstegen, 2014 submitted); thus blood samples can be taken at any time to evaluate circulating AMH. The biological actions of AMH in females, including in humans, is far from being completely understood; but lately, several research groups are focusing their efforts to unravel interesting associations that circulating levels of AMH seem to have with ovarian population, follicular dynamics and ultimately female-fertility. Several reports in veterinary sciences have described that animals with greater ovarian follicular populations clearly have greater circulating AMH. Contradictory, AMH seem to act as a regular of the amount of follicles entering the emergence pool in a follicular wave by inhibiting excessive amounts of follicles from being recruited in a single follicular wave (Monniaux et al., 2014). This is possibly why greater levels of circulating AMH seem to reduce animal sensibility to FSH (Ireland et al., 2007). Thus, AMH might be classified as a regulatory glycoprotein that avoids follicular-reserve in the ovaries from premature depletion. Unlike many reproductive hormones (i.e. estradiol and progesterone), levels of circulating AMH are rather steady throughout the estrous cycle and don’t even seem to be influenced by level of milk production in lactating cows (Souza et al., 2014 – in press). However, AMH is generally much higher in younger males and females, and it decreases with age (Rozner and Verstegen, 2014 submitted). In contrast, nutritional factors such as vitamin-D status in humans (Dennis et al., 2012) as well as negative energy balance in cattle (Souza et al., 2014) seem to have some influence in circulating AMH, although further manipulative studies are urgently needed to draw final conclusions of possible nutritional strategies that may influence circulating AMH. More importantly, AMH will vary greatly across cattle breeds (Batista et al., 2014, Ribeiro et al., 2014) and it definitely needs to be taken into account when defining AMH-thresholds for the differing breeds. Most scientific literature agrees that greater AMH levels are positively correlated with greater follicle counts and, as expected, improved results in terms of numbers of retrieved oocytes during in vitro fertilization (IVF) programs as well as more total structures recovered following superovulation treatments (Monniaux et al., 2010, Guerreiro et al., 2014). However, contrasting results have been found when trying to associate AMH to fertilization and embryo quality in differing animal-species. For example, (Lekamge et al., 2007), utilizing data from 126 women undergoing IVF, found that those with lower AMH had not only less oocytes being produced, but also had decreased fertilization, fewer embryos, and more miscarriages. In contrast, other research groups using cattle as experimental unit even described that greater AMH were associated with increased follicular populations, but surprisingly produced lower quality oocytes after IVF. These findings disagree with a more recent publication using dairy and beef heifers undergoing IVF procedure (Batista et al., 2014, Guerreiro et al., 2014). In addition, results from a recent large study done in superovulated lactating Holstein cows (Souza et al., 2014 – in press) also found a strong correlation between circulating AMH and superovulatory response and total structures collected (Table 1). Interestingly, similarly to results from Guerreiro et al. (2014), greater AMH did not have detrimental effects on embryo quality and/or development as shown in Table 1. 66 67 Using information on circulating levels of Anti-Mullerian hormone (AMH) to enhance embryo production and fertility in cattle (continued) Table 1. Thus, in practical terms to field veterinarians, it appears that AMH can be a reliable marker to predict embryo production potential in cows and heifers undergoing superovulation and IVF routines. The identified threshold for lactating Holstein cows expected to have improved superovulation response was shown to be 123.5 pg/mL. Animals with circulating AMH levels above this threshold should present better superovulatory responses than a random population of cows with unknown AMH levels. QUICK GUIDELINES FOR SAMPLING AND INTERPRETING AMH RESULTS FOR CATTLE Figure 1. Unlike most assay. reproductive hormones that seem to have lowered results with longer blood-storage periods (i.e. progesterone), longer blood sample storage until serum/plasma separation and further analysis will generally yield AMH results that are more variable and with increased readings (Rico et al., 2012). Briefly, it is recommended that blood samples for AMH analysis should be kept at 20 degree Celsius (70 Fahrenheit) or preferably at lower temperatures for no more than 8 hours until serum/plasma separation (Rico et al., 2012); however, Rozner and Verstegen (2014) did not observed any significant differences for samples preserved up to 24 hours before separation. This recommendation then may vary according to commercial assay used. Thus, blood samples for AMH analysis need to be kept refrigerated and processed soon enough mainly to avoid overestimation and greater variability of circulating AMH results. In addition to temperature and time-to-analysis, the type of anticoagulant used in evacuated tubes also affects AMH analysis. Samples collected with evacuated tubes containing EDTA will yield AMH results that are roughly 1.5 times higher than if same samples were collected with evacuated tubes having heparin instead. In addition to these factors, other animal-related variables as previously mentioned such as animal gender, cattle breed, and even age of the animal (as shown in Figure 2 - Rozner and Verstegen, unpublished data) will greatly influence circulating AMH. 68 Figure 2. Therefore, it is highly recommended to strictly follow guidelines prescribed by a specific lab running cattle-AMH assays and avoid mistakes during sampling or data interpretation. AMH AND FERTILITY IN CATTLE Although AMH may serve as a great tool to predict follicular population and, thereby, animal response to IVF and superovulation procedures, its value to predict cattle fertility after artificial insemination (AI) is still under scrutiny. Previous research have shown that cows with lower numbers of antral follicle counts have lower fertility (Mossa et al., 2012). Thus, because circulating AMH is an indirect measure of antral follicle population in the ovaries, it is reasonable to hypothesize that AMH could also be utilized to predict field fertility in cattle. In this regard, Rozner, Souza and Verstegen (2014) have recently studied possible associations between circulating AMH and age at conception in Holstein heifers or interval from calving to conception in lactating Holstein cows, and found nonsignificant relationships between circulating AMH and field-fertility in dairy heifers and mature cows. However, the number of observations was somewhat limited when looking at fertility outcomes and coming from a single herd that exceptionally had animals with greater than expected circulating AMH levels. This database is currently under development to explore possible further associations between AMH levels and fertility in commercial dairy herds. In addition, in beef cattle, AMH has not been related to conception following timed AI protocols in mature Nellore cows (n=296) or heifers (n=643) – data from Dr. Pietro Baruselli’s lab published at 2014 SBTE meeting (Natal, Brazil). It is important though to remember that these results were obtained following timed AI breedings (not natural estrus breedings), which as suggested by (Ribeiro et al., 2014) – timed AI programs may override possible associations between AMH and speed in which cows get pregnant postpartum. In this regard, Ribeiro et al. (2014) have found some positive associations between circulating AMH levels and fertility in dairy cattle as previously suggested (Ireland et al., 2009, Ribeiro et al., 2014). The study by Ribeiro et al. (2014) utilized a large number of dairy cows (n = 1,237) kept in two herds and managed in a 100-d breeding season. Blood sample for AMH analysis was performed 8 days before the beginning of the breeding season, which was assumed to start concomitantly with the 1st AI performed following a synchronization protocol. Then, cows observed in return estrus from 19 to 35 days after first synchronized timed-AI were bred following natural estrus, and finally, bulls were placed with cows on day 36 post-1st AI for 65 more days until the end of the breeding season. Cows were classified into 3 different AMH classes as high (top 20%), average, and low (bottom 20%). Briefly, their results clearly indicate that cows with greater circulating AMH measured just before the start of the breeding season had less embryonic losses to the 1st AI performed following a timed AI protocol. Moreover, cows that did not conceive after the 1st AI became pregnant in a faster pace throughout the breeding season as compared to cows with lower AMH. Further studies are obviously needed to confirm these promising results, but it appears that circulating AMH might also be helpful to guide breeding strategies in cattle. Despite the debate on the possibility of identifying cattle with improved fertility; because number of ovarian antral follicles is correlated in cattle dam-daughters (Walsh et al., 2014), we have also tried to look into possible associations between circulating AMH in dam-daughter pairs (Figure 4). The correlation in circulating AMH in dam-daughter pairs although significant, it was lower than we expected (r = 0.18), but might allow for selection for improved fertility in cattle. The repeatability of the AMH assay makes it a valuable practical method that can be readily used in the field to improve the efficiency of reproductive technologies commonly used in cattle.
