Summary
Neogen’s Benzodiazepine Group ELISA kit provides ultra
sensitive detection of over 20 drug analytes in a variety of
matrices. Designer benzodiazepines are increasingly widespread
throughout Europe as recreational drugs and non-prescription
medication. Our test kit is successful in the detection of both
traditional and synthetic benzodiazepines and is intended for
forensic use only. It is recommended that all questioned
samples be confirmed by a quantitative method such as gas
chromatography/mass spectrometry (GC/MS).

Summary of Protocol
- Add 10 µL samples or calibrators
- Add 100 µL ready-to-use enzyme conjugate solution
- 45 minute incubation at room temperature
- Wash plate 3 times
- Add 100 µL TMB Substrate
- 30 minute incubation at room temperature
- Add 100 µL Stop Solution
- Measure OD at 450 nm
Sample Dilutions
The recommended minimum
sample dilution is 1:10 for urine
and 1:5 for whole blood
and oral fluid.

β-Glucuronidase Treatment of Samples
If detection of total benzodiazepines including the glucuronide
metabolites is desired, an initial sample treatment with a
hydrolysis enzyme will be necessary.
Neogen's β-Glucuronidase treatment will hydrolyze the
glucuronic acid from lorazepam glucuronide to yield
unconjugated lorazepam for better detection of total
lorazepam in Neogen’s Benzodiazepine Group ELISA Kit (see
figure below)*.

*The ββ-Glucuronidase sample treatment for lorazepam glucuronide should be able to be utilised for
other glucuronide metabolites of other drugs. Neogen's β-Glucuronidase sample treatment has only
been tested on lorazepam glucuronide and Neogen’s Benzodiazepine Group ELISA Kit. It has NOT been
tested on any other glucuronide metabolites and/or forensic detection kits.
