"Markers of Sperm Cryopreservation Success" IRB #42890 (PI) June 20, 2012- June 19, 2014. In collaboration with
Dr. Thomas Walsh and resident physicians, we investigate predictors of success of sperm freezing. We will
identify sperm measures (including motility, kinematics, total numbers, morphology), semen analysis
measures, processing variations, and clinical diagnoses that may predict the recovery of motile sperm postthaw and the usefullness of thawed sperm for reproductive purposes.
"Physiology of Ejaculated Human Sperm" IRB # 06-0264 (J. Amory, PI; C. Muller Co-Investigator), March 2009 present. Human male subjects are recruited by Dr. Amory or by Dr. Muller through the Male Fertility Lab
(for infertile men), and screened for health, reproductive hormones, and semen analysis. Subsequently,
semen samples from consented donors are used by co-investigators. In Muller's lab, sperm function tests
(e.g., acrosome reaction, hyperactivation), effects of stimulators on purified sperm, effects of contraceptive
agents, and DNA fragmentation are studied. Semen and sperm samples are processed for analysis of
retinoic acid by collaborator Dr. Nina Isoherranen. In collaboration with Dr. Kim Woodrow in
Bioengineering, sperm are used in development of novel contraceptive methods. In collaboration with Dr.
Jeffrey Riffell in Biology, sperm chemotaxis is studied using laminar flow devices and automated sperm
kinematic analysis in Muller's lab. In collaboration with Dr. Margaret Shnorhavorian of Seattle Childrens
Hospital, sperm from survivors of childhood cancer are analyzed. In collaboration with Dr. Michael
Skinner of Washington State University, samples are collected and processed by Muller's lab for analysis of
the human sperm epigenome.
"Intrauterine Insemination for Couples with HIV Discordancy" IRB # 37365 (B. Houmard, PI) June 2009-2013. In
this study, we investigate the most efficient laboratory method for removing HIV virus particles or cells
expressing HIV from spiked semen samples. Second, with further IRB approval, we use this approach in a
clinical setting for HIV discordant couples seeking a pregnancy where the man is HIV-positive. The Male
Fertility Lab processes the sperm samples for this study.
“Human Testis Analysis” IRB # 38286, (Muller, PI) April 2010- present. Fixed and sectioned normal (noncancerous) human testes from men with various diagnoses are analyzed for spermatogenesis by
morphological and molecular markers of Sertoli cell and germ cell function. Comparisons are made, where
possible, with semen analysis and reproductive and medical histories. Samples are obtained from Pathology
files, and analyzed by Drs. Muller, Walsh and Amory.
"Retinoic Acid and Spermatogenesis in Man" IRB # 37399 (Amory, PI; Muller Co-Investigator). 2010-present.
Retinoic acid and its metabolites are measured in semen, sperm, and testicular tissue obtained by biopsy
from men with various fertility diagnoses. The Male Fertility Lab / Andrology Research Lab processes the
sperm samples for this study.
"DNA Fragmentation, Oxidative Stress and Pro-Inflammatory Cytokines in CP/CPPS" IRB # 27656 (Muller, PI).
2006-1/5/2014 (continuation for data analysis). Chronic pelvic pain in men, formerly known as “prostatitis”
is a common complaint. We thought that some of this pain might be due to oxidative stress and subsequent
damage to sensory nerves. We hypothesized that such oxidative stress would be manifested by oxidative
damage to the men’s sperm cells, which accumulate and do not repair DNA damage. We measured sperm
DNA damage, reactive oxygen species and anti-oxidants in semen of pain and control subjects. We found
that, on average, the controls had higher levels of DNA damage and reactive oxygen activity than the pain
subjects. However, pain subjects had lower levels of the anti-oxidant catalase in semen. Pain subjects also
had more white blood cells, higher nitric oxide levels, and higher levels of the chemokine MCP-1. Thus,
our original basic hypothesis is not supported, but several details are worth further investigation. Research
protocols have been completed by the Andrology Research Lab. We are still analyzing data.

"Predictive markers of sperm cryopreservation success in men with cancer" IRB # 36608 (A. Jaime; C Muller, PI
Faculty Sponsor and Co-Investigator), June 2009-Dec 2011. The objective of this research project is to
investigate the sperm dysfunctions of men diagnosed with various types of cancer who are undergoing
radiation and/or chemotherapy, before, during and after cryopreservation at the Male Fertility Lab. Semen
analysis data and data from cryopreservation cycles pre- and post-thaw are used to investigate the
relationships among types of cancer and sperm function and cryosurvival.
