Morpheus® II integrates reagents that are not seen in other screens,
especially less common additives. As a consequence, the screen
should have an impact not only on crystallisation but also on the
overall structure determination process.
It follows the general design principles of the original Morpheus
screen, but less typical additives have been included, such as heavy
metals, NDSBs, polyamines, amino acids and monosaccharides. In
addition, various polyols have been added for cryoprotection along
with innovative buffer systems.

96-CONDITION ADDITIVE SCREEN WHICH CONTAINS A
UNIQUE RANGE OF LIGANDS, HEAVY ATOMS AND POLYOLS.
Morpheus® Additive Screen uses reagents that are not seen in
other screens, especially less common additives. Some additives
such as metal divalent cations can induce new crystal contacts.
Heavy atoms were integrated to help with ab initio structure
determination. Most of the other additives are included to
enhance protein stability and solubility (carboxylic acids,
polyamines and monosaccharides).

n Targeted incorporation of 35 low-molecular weight PDB ligands into
96 conditions.

amino acids and monosaccharides to modify the solubility and
stability of many proteins.
n Incorporation of heavy atoms as additives for experimental phasing.
n Also ideal for membrane proteins as contains PEGS and low salt.
n The inclusion of Non-detergent sulfobetaines (NDSBs), polyamines,
amino acids and monosaccharides to enhance solubility and stability
of many proteins for crystallization.
n Contains heavy atoms which can be used for experimental phasing.

The term "shotgun screening" was coined early in the Structural
Genomics era and refers to the process of setting up
experiments using pre-mixed cocktails until a crystal of
sufficient quality is obtained.
The Wizard Classic reagents are proven to be a highly effective
starting point in the screening of biological macromolecules. The
Wizard Classic formulations include a large range of crystallants,
buffers, and salts covering a broad range of crystallization space
at pH levels from pH 4.5 to pH 10.5.
The best place to start screening is within the context of
previously successful crystallization space. "Although only 14%
of successful crystallization conditions from the PDB are
identical to a commercial condition, almost 40% of the PDB
conditions can be obtained by trivial optimization of a
commercial cocktail.” (Fazio et al) So this is a reasonable place
to commence screening.

Crystals can be frozen directly
from their growth drops, avoiding
the additional step of preequilibration with an artificial
cryo-condition that can damage
the crystal. Eleven different
cryocrystallants and sparing use
of glycerol ensures a broad
sampling of possible cryo
conditions.
Researchers from the Joint
Center for Structural Genomics
(JCSG) formulated this screen
with the goal of maximizing the
coverage of the crystallization
parameter space (buffers, salts,
precipitants, and pH) and
eliminating redundancy. The
buffers give final solution pHs
between 4.2 and 10.5.
Precipitants include volatile
reagents such as ethanol, high
molecular weight polymers such as
PEG-8000, and salts such as
ammonium sulfate.

This semi-systematic screen has been designed to work in synergy
with Lipidic Cubic Phase (LCP), now commonly used in membrane
protein crystallization, without perturbing phase formation.

membrane protein crystal structures has

Molecular Dimensions provides the largest

crystal growth data mined from the most

of the first specialised additive screen. For in
meso methods a Sponge Phase screen, and

DIACON™
DIACON IS IDEAL FOR GENTLE AND EFFICIENT
BUFFER EXCHANGE, DESALTING AND LIGAND
EXCHANGE OF PROTEIN FROM LOW VOLUME
SOLUTIONS OF UP TO 0.5ML. THE SAMPLE
SOLUTION IS MANUALLY PIPETTED INTO THE
DEVICE WHICH IS THEN HELD UPRIGHT IN
THE DIALYSATE VIA THE FLOATING RACK
PROVIDED, ENSURING THE MEMBRANE IS
ALWAYS IN CONTACT WITH THE DIALYSATE.
The volume of dialysate can be as large as desired; the larger the
volume the larger the concentration differential and therefore a
faster rate of diffusion occurs, optimizing dialysis. Dialysis is
recommended for 2-3 hours and then change the dialysis buffer
leaving for another 2-3 hours. A third dialysis can be carried out if
necessary. Diacon employs specially selected regenerated
cellulose membranes.

DIATUBE
™
Diatube holds up to a 20 mL sample solution one side of the dialysis
membrane and up to 25 mL dialysate on the other, creating a
concentration differential. Diatube is ideal for buffer exchange and
desalting of sample volumes from 5 to 20 mL. The dialysate can be
changed at any point to recreate the concentration differential for
the dialysis process to continue.
The disposable Diaplate is a 96 well micro-dialysis plate for very
small volumes of up to a maximum of 3.2 µL. The sample solution is
pipetted onto each of the 96 membranes with up to 0.35 mL unique
dialysis solutions in the wells used for exchange. Dialysis is
recommended for 2-3 hours and then the dialysis buffer is changed,
left for another 2-3 hours to achieve over 90% exchange. Further
dialysis steps can be carried out as required. Incubation on a rocking
table for speed and a moisture chamber for full humidification can
be applied to the experiment if desired. The Diaplate™ kit includes:
Diaplate with 200 Micron Pressure Adhesive Spacer, 200 Micron
UVP Cover Film, engraved UV Protective Cover and a sealing Paddle.

Owing to the high position of the membrane and the UV transparent
cover, samples on Diaplate can be observed by microscopy,
fluorimetry, turbidimetry and light scattering. Diaplate is therefore
ideal for crystallization by dialysis by changing ionic strength, or

suitable for ligand exchange, slow controlled addition of
cryoprotectant and for protein stability screening.

Uricon is a static concentrator used for the
rapid concentration of protein solutions.
Up to 15 mL sample solution is pipetted
into the isolated chamber within the device
where one inner surface is a membrane of

backed by a pressure resistant absorbent
pad and gel that pull the solvent and salts
through the ultrafilter by absorption,
concentrating the sample. Uricon has an
impermeable dead stop at the base of the

There has been some speculation that water vapour may evaporate
through a thin film cover. As yet there is no experimental evidence
to substantiate this suggestion. Users have also speculated that it
may be related to how well the film is pressed down onto the
adhesive spacer.
Crystal growth plate design has evolved considerably
over the past decade to incorporate excellent optical
properties for imaging experiments; plastic surfaces to
address drop behaviour; birefringence; hydrophobicity;
UV transparency and well geometry for ease of
harvesting crystals. Molecular Dimensions high quality
moulded plates, manufactured by Swissci, have no flow
lines and no bubbles and feature all of the above
qualities for sitting drop, hanging drop, microbatch,
counter diffusion and versatile sandwich devices for LCP.

CONTENTS OF THE STARTER KIT
7 x Uni-Pucks
The Universal V1-Puck (uni-puck) is a sample pin storage and
shipping container compatible with the majority automated sample
mounting systems worldwide. Each Uni-puck holds 16 standard
18 mm sample pins in CryoCaps. It consists of two parts: 1) the
sample enclosure, and 2) the sample base piece. Each part of the
Uni-puck has a unique serial number for identification. Custom
Colours and Serial Numbers are available upon request. (Always
consult your local beamline user support contact about specifics of
container and sample pin compatibility).
1 x Puck-Shelved Shipping Cane
Seven uni-pucks may be shipped or stored in this cane, which fits
into the CX100 transport Dewar (Dry Shipper - MD7-21).
1 x Angled Cryo-Tongs
These tongs are designed specifically to hold and transfer cryocooled pucks.
1 x Puck Separator Tools
This set of tools includes a base removal platform and a porcupine
push tool to recover your samples.
1 x Puck Wand
Magnetic puck-base holder; it is designed for attaching the base
plate to the sample enclosure.
1 x Puck Dewar Loading Tool Set

Features of CryoSol:
Blue polypropylene container with handle and lid 1 L
n Use in co-crystallization screening of protein-ligand complexes.
This represents the complete answer in low-cost, high efficiency
Dewar flasks. Utilising a proven borosilicate glass inner vessel,
MD7-807 features a tough, blue plastic, shock-absorbing
container with many safety features. The unit is easily dismantled
to facilitate refill changes and includes a coated handle and a
'push-fit' insulated lid, vented for safe handling of all liquid gases.
n Preparation of ligand-loaded cryosolutions for soaking experiments.
n Ideal for the solubilization of hydrophobic ligands.
n Protein friendly mixes of DMSO/dioxane/ethylene glycol should
improve ligand solubilization.
n Contains 2,3-butanediol- compatible with enzymatic activity.
n Mixed solutions can be used as an additive screen (up to 5% in
precipitant).

Fitted in highest-quality stainless steel containers, therefore rustfree, anti-corrosive, all hygienic and strong. These Dewars have an
all stainless steel lid with self-contained insulation. Toggle-clamps
secure the lid to the body for safe transportation.

sugars were solubilizing the complex too much so Dioxane was
added to the PEG to crystallize it.
