THE PURPOSE OF The G&P Foundation for
Cancer Research is to encourage the development of
more effective therapies for
patients with leukemia, lymphoma and related cancers. The Foundation funds innovative
clinical or basic science research that will lead
to novel therapeutic approaches that could
replace, or be used in combination with existing
effective therapies. Such therapeutic approaches
could include integrative (integrative/complementary) medicine.
The Foundation funds research to improve the
efficacy of cancer treatments, reduce their toxicity and improve the quality of life of patients
with leukemia or lymphoma.
The Foundation also supports limited scientific
interchange between select leaders in different
areas of bio and integrative medical research, to
develop unique and important collaborative efforts
that will lead to improvements in the treatment
and care of patients with leukemia and lymphoma.

A STAR-STUDDED ARRAY OF CELEBRITIES joined Grammy-nominated songwriter and
G&P Co-Founder Denise Rich at the New York Marriott Marquis for the fourth Angel
Ball gala, helping to raise $3.3 million for The G&P Foundation for Cancer Research.
The black-tie benefit, which featured a cocktail reception, dinner and live auction,
recognized four honorees for their outstanding contributions as humanitarians and
philanthropic leaders: singer/songwriter Patti LaBelle; Russell Simmons, Chairman

heartfelt rendition of “Somewhere Over the Rainbow”, and was joined by Stevie
Wonder at the piano, Natalie Cole and Wonder’s daughter, Aisha Morris. The night
culminated with all of the performers uniting onstage to sing the Martha Reeves
classic “Dancin’ in the Streets.”

Third in a Series
IN OUR CONTINUING EFFORT to introduce
readers to the esteemed physicians on The
G&P Foundation Medical Advisory Board,
Inspirations presents its series of interviews
showcasing a member of our board in their
own words.
In this third installment, we are proud to
introduce you to Janet Davison Rowley, M. D.
of The University of Chicago. Dr. Rowley is
the Blum-Riese Distinguished Service
Professor in the Departments of Medicine,
in Molecular Genetics and Cell Biology and
in Human Genetics. She received her
medical degree from the University of
Chicago in 1948. She joined the research
faculty at the University of Chicago in 1962
and became a Professor in 1977. Dr. Rowley
has contributed significantly to advances in
understanding of genetic changes in cancer.
Her discoveries have revolutionized the view
of hematologists/oncologists and cancer
biologists regarding the critical importance
of recurring chromosome abnormalities in
cancer cells.
She has published over 400 papers and book
chapters describing her research in
leukemia and related areas and has received
numerous awards including Dameshek Prize
(1982), Medal of Honor (ACS, 1996), and more
recently the Dorothy P. Landon Award
(2005). She is a member of the National
Academy of Sciences (1984), Institute of
Medicine (1985), American Academy of Arts
and Sciences (1991), and American
Philosophical Society (1993). She has
received nine honorary degrees including
one from Oxford in June 2000 and from
Lund in 2003.

“I should begin by saying how important I
think the work of the Foundation is,
especially today as budgets for science are
being constrained by other federal
requirements.
The goal of The G&P
Foundation namely to fund young scientists
and physicians and others involved in the
care of patients with cancer is more needed
than ever. Young scientists have a very hard
time breaking into the system and getting
funding for starting their laboratories. I was
very fortunate back in 1962 when I began in
research to be part of a project that was
funded by the national government so I
didn’t have to apply for grants in the first
few years.
The project that I have been involved in
for more than thirty years is the study of
genetic changes in leukemia cells and
lymphomas. The tools I used to study
these genetic changes in the malignant
cells were chromosomes. The normal
human cell has 46 chromosomes of particular sizes and shapes. What has become
apparent over the course of my studies is
that many leukemia cells
have altered chromosome numbers (too many
or too few) or they have
altered chromosome sizes
and shapes due to breaks
in chromosomes and
rearrangements.
I was very fortunate in
1972 to be the first to
identify the chromosomes involved in two of
these re-arrangements,
called “translocations”.
In cancer and especially
leukemia and lymphoma,
chromosomes are broken
in specific places, and
their ends are swapped
so that different size pieces of the chromosomes are exchanged. This leads to
identifiable changes in the chromosomes
when you look at them under the microscope.
The most well known translocation I discovered was one involving chromosomes
9 and 22. It had been known since 1960
that a particular form of leukemia called
Chronic Myeloid Leukemia (CML) had a
unique small chromosome. Because it
was discovered in the city of Philadelphia
it was called the Philadelphia or the Ph
chromosome. With the staining techniques that were used at the time, chromosomes all stained uniformly. They
could identify this very small marker
chromosome but how it came to be wasn’t
clear. I applied a new technique that
showed that each chromosome pair had a
unique pattern of alternating dark and
light stripes to study this mystery marker
chromosome. I could see that in fact the
Ph chromosome was actually a translocation where a large piece of chromosome
22 went to the end of chromosome 9 and a
smaller piece of chromosome 9 went to
chromosome 22 giving rise to this small
Ph chromosome.
The importance of this is that other laboratories analyzed the DNA at the sites of
the chromosome breaks and discovered
that the gene on chromosome 9 was the
Abelson gene, which had already been
identified in a mouse leukemia. The consequence of the translocation was to
break within the gene and to move part of
the Abelson gene to chromosome 22, next
to a gene called BCR that
is also broken. So you
end up with a new,
unique fusion gene on
the Ph chromosome,
which consists of part of
the normal BCR gene on
22, fused to the back half,
if you will, of the
Abelson gene, which is
normally on chromosome 9.
This fusion leads to the
activation of the Abelson
gene whose function is
as a tyrosine kinase. The
tyrosine kinases act to
increase cell growth.
The fusion then leads
these cells to grow not necessarily more
rapidly but it keeps them from stopping
growth when there is sufficient number of
white blood cells. This, in turn, with
other genetic changes whose identity is
unknown at the present time, leads the
cell to become a leukemic cell.
This discovery has turned out to be
extremely important because others, most
especially Brian Druker at Oregon Health
& Science University, working with a
chemist at Ciba, identified the compound
known as STI 571 that inhibited the
growth of cells with this abnormal
Abelson fusion protein. This drug was
then named Gleevec, now Imatinib, which
has transformed the treatment of patients
who have CML.
The challenge for the future is to identify
the same kind of drug to treat patients who
have acute forms of leukemia, to treat them
more effectively. This is especially urgent for
patients who have Acute Myelogenous
Leukemia (AML) who are older, i.e. over the
age of 60, because the outcome of
treatment in these patients is still dismal.
So, there is need for continued research to
find additional drugs to treat these other
forms of leukemia.”
