Biennial Report 2014-2015
The Parkinson’s Disease Foundation® (PDF®) is a leading
national presence in Parkinson’s disease research, education
and public advocacy. We are working for the nearly one
million people in the US who live with Parkinson’s by funding
promising scientific research while supporting people living
with Parkinson’s through educational programs and services.
Since its founding in 1957, PDF has dedicated nearly $110
million to fund the work of leading scientists throughout the
world and nearly $47 million to support national education
and advocacy initiatives.

Dear Friend of the Parkinson’s Disease Foundation:
It is our pleasure to be sharing with you this report from the Parkinson’s
Disease Foundation (PDF) on the work we have been able to accomplish
with your support during fiscal years 2014 and 2015.
During this period, the environment for a nonprofit research and advocacy
organization like ours has been challenging. Federal support for Parkinson’s
disease science has been in short supply, meaning that fewer research
proposals are being funded (just 14 percent of proposals to the National
Institutes of Health received support during the period of this report, down
from 25 percent just a few years earlier). Meanwhile, in the private sector,
according to a recent report by The Institute of Medicine, six of the top 10
pharmaceutical companies have reduced their investments in brain science,
by an average of 25 percent.
In this environment, it is even more important than usual that PDF create a
strategic focus that moves the needle on Parkinson’s research once and
for all. We call it, “Progress, Passion and Impact.” It means progress, in
combining our commitment to tried and true formulas for success with a
promise to test new ideas. It means passion, in the excitement of the young
scientists whose work we fund; in the energy that we see among our patient
leaders; and in the devotion of our Board and staff to their work. And it means
impact, in the questions that we ask of every PDF program. How will it
advance science? How will it enhance lives, and how many? And how will
it fuel hope, for improved care and an accelerated cure?
To give content to these ideas, PDF initiated in 2014, under the guidance of
our Board of Directors, Scientific Advisory Board and People with Parkinson’s
Advisory Council, a year-long examination of how far we have come, and how
we can contribute to solving the Parkinson’s puzzle moving forward. PDF’s
strategy is to build a team of people — the best and brightest leaders in
science, health care and the patient community — and then mobilize them
to work towards the cure together.

This team of leaders propels the progress of our field. They all harness
the passion of our players. And they all hold the promise of impact upon
the problems and challenges that engage all of us in the Parkinson’s
community. Here are three examples of new initiatives from the past
two years that vividly illustrate these values:
 Created npj: Parkinson’s Disease, a new online, public access journal
of Parkinson’s disease science in association with Nature Publishing
Group, the premier publisher of scientific journals in the world;
 Initiated a partnership with The Edmond J. Safra Foundation to
expand the Edmond J. Safra Foundation Visiting Nurse Faculty
Program at PDF, which has trained 150 faculty members at
nursing schools nationwide;
 Introduced a series of Parkinson’s Advocates in Research Leadership
Awards, providing the first funding opportunity for scientists to
expand their research teams to include people living with Parkinson’s.
In different ways, these initiatives bring PDF’s program imagination and
experience together with its institutional relationships to leverage small
investments into large results.
It remains for us to wish you a happy and healthy start to 2016, and to
thank you for all you have done over the years to support and make
possible the work of the Parkinson’s Disease Foundation. We simply
could not do it without you, and we hope that you are as proud of your

Program Highlights
Tim Bartels, M.Sc., Ph.D., of Harvard Medical School
and Brigham and Women’s Hospital, a 2012 International Research
Grants Program awardee, was part of a team that shed light on
how and why alpha-synuclein (α-synuclein), a protein found in all
brain cells, accumulates in Parkinson’s disease (PD). According to
their research, α-synuclein normally occurs as a group of four
strands, called a tetramer, and a change in this structure results
in α-synuclein clumping together. The study suggests that by
stabilizing α-synuclein’s shape to remain as a tetramer, the
accumulation of this protein may be prevented.
Sarah Ozinga of Cleveland State University
and Cleveland Clinic, a 2013 PDF Summer Fellow,
was part of a team that concluded that the
iPad tablet offers a reliable and affordable way
to assess and monitor balance difficulties in
people with PD. Since postural instability
remains one of the most difficult to treat aspects
of Parkinson’s, there is hope that consumer
electronics can help clinicians to develop better
ways to treat and manage these symptoms.
David Sulzer, Ph.D., of the PDF Research
Aiqun Li, Ph.D., of the New York Stem Cell Foundation, a
2014 Lucien Côté Early Investigator awardee, and colleagues
created induced pluripotent stem (iPS) cells from skin samples
from a pair of identical twins, both with a glucocerebrosidase
(GBA) genetic mutation that increases risk of PD, but only one
twin had the disease. Researchers looked for differences
between the twins’ neurons and found that while the neurons
generated from both twins produced less dopamine (the chemical
that is deficient in Parkinson’s disease) than
normal, the twin with PD had even lower levels
of dopamine because he had higher amounts

brain cells communicate in Parkinson’s rather than from a “toxic”
effect of taking levodopa, the most common PD medication.
Their research found that as PD progresses and more dopamine
is lost, remaining brain cells become oversensitive to dopamine.
The study supports an emerging view that PD progression, rather
than length of time taking levodopa, underlies dyskinesias.

Parkinson’s Advocates in Research (PAIR) Leadership awardees,
partnered to study how building patient-researcher collaborations
can accelerate Parkinson’s research. Dr. Kluger leveraged this
support to attain a $1.9 million dollar contract from the Patient-

Centered Outcomes Research Institute (PCORI) to examine if outpatient palliative care improves patient-centered outcomes in PD.
Jennifer Goldman, M.D., of the
PDF Research Center at Rush University, used
research
MRI to identify distinct structural differences in
the brain that are associated with hallucinations
in Parkinson’s. These differences occur in the
areas of the brain associated with visual
perception, and help explain why most hallucinations in PD are
visual in nature (rather than auditory or tactile). This information
may allow earlier detection of individuals at risk for hallucinations
in PD and ultimately, the development of therapies targeting
hallucinations and other impairments in visual perception.
The Community Choice Research Awards
were launched in 2013, inviting the Parkinson’s community for
the first time to identify and set research priorities to be
addressed by multidisciplinary teams (including people with
Parkinson’s). In 2014, PDF announced $30,000 in funding to
address two disabling, yet under-recognized symptoms of
Parkinson’s — fatigue and gastrointestinal dysfunction — identified as priorities through participants in this initiative.

The PAIR Program
expanded to provide training to more than 260
Research Advocates who have worked with more than 400 research professionals
and have helped enroll 1,200 people in clinical trials testing new treatments. PDF
Research Advocates are involved in all stages of the research process — advising
the Food and Drug Administration (FDA) on patient needs and priorities; collaborating as co-investigators and steering committee members of local and national
research projects funded by the Patient-Centered Outcomes Research Institute
(PCORI) and the National Institutes of Neurological Diseases and Stroke (NINDS);
and advising pharmaceutical companies on study development and implementation.

PDF’s National HelpLine, available
in over 200 languages, answered more than 11,000 questions about Parkinson’s
and connected people to thousands of community resources.
PD ExpertBriefings, PDF’s online educational initiative, expanded to include
over 40 web-based programs that offer people living with Parkinson’s and health
professionals tools to better understand the disease and treat it effectively. Since
2013, more than 2,000 health professionals and 20,000 people with PD and care
partners from 43 countries have participated in our online programs.
