Immersion OIL Rules: Always keep immersion oil bottle CLOSED. This prevents water evaporation and maintains the integrity of the oil Keep oil bottle CLEAN. Oil should never be found along the sides of the bottle or around the neck Apply only a SMALL AMOUNT of oil to your coverslip Oil droplet Coverslip Always load your sample on the microscope stage with the STAGE LOWERED to the lowest focus position Always REMOVE OIL using dry Lens Paper from the objective when changing slides. Re-apply a small amount of new oil to the new slide If you are imaging a sample using an oil objective, do not toggle between objectives. Image with that objective only! When you are done your session, clean objective using Dry Lens Paper Do not get oil on any air objectives! Attention Users!! FluoArc Mercury Lamp: The FluoArc lamp must be on for at least 30 minutes before being turned off AND must not be turned on again within 1 hour of being turned off. 1. When starting, before you turn on the FluoArc lamp, make sure it has been OFF for at least 1 hour from previous use. 2. When finishing, before you turn off the FluoArc lamp, make sure it has been ON for at least 30 minutes. Objectives: If you use oil on an oil-objective, please wipe it off with dry Lens Paper after you have finished. ApoTome: Before toggling ApoTome power box switch off, make sure that ApoTome slider is pulled out to click stop position 1 Your Data: You can temporarily save your data under My Documents, your lab-folder, your folder You are responsible for backing-up your own data at all times. Do not rely on this computer as storage for your data. Once you have backed-up your files elsewhere, please delete your files from this computer. Attention Users!! Log Book: All users must log the time spent on the station as well as the time the fluorescence lamp was turned on/off in the log book. Guest Users: Others and guest users can (with permission) use this microscope on Mondays and Thursdays only. Computer: This computer is to be used only for image acquisition. No other work is permitted!! Zeiss Imaging System Help Line: 1-800-509-3905 Zeiss Axio Imager Z1 Start Up Procedure 1. Remove dust cover from microscope 2. Toggle FluoArc lamp switch ON 3. Toggle yellow power bar switch ON 4. Toggle black power bar switch ON 5. Toggle Power Supply 230 box switch ON 6. If using ApoTome, toggle ApoTome box switch ON 7. Turn on computer 8. Log in: - User Name: GuestUser - Password: -leave blank- 9. Open Program: AxioVision Shut Down Procedure 1. If used oil, gently remove oil from lens using neatly folded dry lens paper 2. Lower stage and put 5x objective in place 3. Close AxioVision program 4. Shut down computer. Wait for computer to completely turn off 5. If used ApoTome, move slider out to click stop position1, then toggle ApoTome box switch OFF 6. Toggle Power Supply 230 box switch OFF 7. Toggle black power bar switch OFF 8. Toggle yellow power bar switch OFF 9. FluoArc Lamp: - Regular work hours: Set intensity to 50% on control pad (F2), and leave lamp ON - Non-work hours: Turn OFF lamp when you are finished only if no one else is using it after you and if the lamp has been on for at least 30 minutes Cover microscope with dust cover, but DON’T COVER THE HOT LAMP HOUSING!!
