Spin–Mix–Spin technology for 2 PCR plates at the same time, thus saving time

Rapid PNGase F
PNGase F is the most effective enzymatic method
for removing N-linked oligosaccharides from glycoproteins. PNGase F digestion deaminates the
asparagine residue to aspartic acid, and leaves the
oligosaccharide intact for further analysis. Rapid
PNGase F is an improved reagent that allows the
complete and rapid deglycosylation of antibodies
and immunoglobulin fusion proteins, as well as other
glycoproteins, in minutes. All N-glycans are released
rapidly and without bias, and are ready to be
prepared for downstream chromotography or mass
spectrometry analysis.

NEB® Golden Gate Assembly Mix
The efficient and seamless assembly of DNA fragments, commonly referred to as Golden Gate assembly, has its origins in
1996 when, for the first time, it was shown that multiple inserts
could be assembled into a vector backbone using only the
sequential or simultaneous activities of a single type IIS
restriction enzyme and T4 DNA Ligase. This method can be
accomplished using Type IIS restriction enzymes, such as BsaI,
and can also be used for the cloning of single inserts. The
method is efficient and can be completed in one tube in as little
as 5 minutes for single inserts, or can utilize cycling steps for
multiple inserts. Golden Gate Assembly has been widely used
in the construction of custom-specific TALENs for in vivo gene
editing, among other applications.

NEB Golden Gate Assembly Mix
In its simplest form, Golden Gate Assembly requires a Type IIS
recognition site, in this case, BsaI (GGTCTC), added to both ends of
a dsDNA fragment. After digestion, these sites are left behind, with
each fragment bearing the designed 4-base overhangs that direct
the assembly.
HiScribe™ T7 ARCA mRNA Kits
Most eukaryotic mRNAs require a 7-methyl guanosine (m7G)
cap structure at the 5 ́ end and a poly(A) tail at the 3 ́end to
be efficiently translated. The HiScribe T7 ARCA mRNA Synthesis
Kit (NEB #E2060) is designed to synthesize capped and tailed
mRNAs for variety of applications. Capped mRNAs are
synthesized by co-transcriptional incorporation of Anti-Reverse
Cap Analog (ARCA) using T7 RNA Polymerase. A poly(A) tail is
then added by E. coli Poly(A) Polymerase. A separate version
of the kit, without E. coli Poly(A) Polymerase (NEB #E2065), is
available for use with DNA templates encoding a poly(A) stretch
or not requiring a poly(A) tail. The kits also include DNase I and
LiCl for DNA template removal and quick mRNA purification.

Luciferase expression in U2OS cells. Purified Cypridina luciferase
RNA produced as indicated was co-transfected into U2OS cells
with purified Gaussia luciferase mRNA. mRNAs produced using the
HiScribe T7 ARCA mRNA Kit (with Tailing) are 5 ́-capped and have
3 ́ poly(A) tails. After 16 hours incubation at 37°C, cell culture
supernatants from each well were assayed for CLuc and GLuc
activity. Luminescence values were recorded and used to calculate
relative luciferase activity.

Highly specific and sensitive nanoimmunoassay of PARP1
A toolkit to measure cell death
Background
PARP1 is the primary member of the poly(ADP-ribose) polymerase family, whose function is to signal DNA damage (and to
recruit repair proteins) by PARylation. PARP1 is also involved in
multiple cell death pathways, including apoptosis, necroptosis,
autophagy, and a relatively new pathway termed parthanatos.
It has been implicated in a new form of cell death termed
parthanatos. PARP1 can also promote tissue survival by shifting
the balance of cell death programs between autophagy and
necrosis. Clinical studies have shown vulnerability to PARP
inhibitors in DNA repair defective cancers. Anti-PARP1 (internal)
