Guidelines for Using Lyophilized TaqMan® Assays with Master Mix
For 7500/7300 System users
To run an experiment on your real-time PCR instrument using strip-tubes, one of the
following precision holders is required.

Steps:
1. Open the real-time PCR instrument tray and remove the “Precision Plate Holder
for plates” from the instrument tray if necessary.
2. Place the “Precision Plate Holder for tubes and strips” (refer to Table 1) in the
instrument tray as shown in Figure 1.*
Step 1: Open tray and remove holder for plates.
Step 2: Insert the holder for strip-tubes.

“Lyophilized TaqMan® Gene Expression Assays with Master Mix” experiment protocol

For StepOne™ and StepOnePlus™ System users
To run an experiment on your real-time PCR instrument using strip-tubes, one of the
following adaptor trays is needed.

Use only Fast consumables (reaction plates, strip-tubes, and tubes) with the StepOne™
and StepOnePlus™ Systems.
Steps:
1. Open the real-time PCR instrument tray.
2. If you have been running plates, place the adaptor tray (refer to Table 2) directly
on top of the tray of the real-time PCR instrument in the correct orientation (A1
well position on the top left) as noted in Figure 2.

Step 2: Place strip-tube adaptor directly on top.

For ViiA™ 7 System users
To run an experiment on the ViiA™ 7 Real-Time PCR instrument using strip-tubes, the
following trays and adaptors are needed. You can find product details for the Retainer
Trays on our website, while ViiA™ 7 adaptors are only bundled with the ViiA™ 7
instruments.

Steps:
1. Touch
to eject the tray arm.
2. When the tray arm opens, pull the latch, then lift and remove the plate adaptor
(Figure 3, Steps 2 and 3).
3. Replace with the appropriate Tube Adaptor (see Table 3).
4. Cap and place the strip-tubes in the corresponding Retainer Tray (Table 3) and
place the Retainer Tray with strip-tubes on the Tube Adaptor (See “Loading
0.2mL Strip-Tubes on the Retainer Tray” section on the following page).
5. Make sure “A1” of the Tube Adaptor aligns with “A1” of the Retainer Tray.
6. Touch
again to close the tray arm.
Steps 2 & 3: Replace the Plate Adaptor

Loading 0.2mL Strip-Tubes on the Retainer Tray
The Retainer Tray for Standard 96-well (0.2mL) tubes and strips (Part No. 4381850) is
comprised of two parts; a top unit and a bottom unit.

Snap open the top unit of the Retainer Tray (Figure 4, Step 1).
Put the un-capped 0.2mL strip-tubes into the bottom unit of the retainer tray.
Snap the top unit of the retainer tray back on top (over the un-capped strip-tubes).
Cap the strip-tubes, the optical strip-tube caps should be on top of the top unit of
the Retainer Tray.
Step 1: Snap open the top unit of the Retainer
Tray.
Steps 2 to 4: Assemble the tube strip with
the Retainer Tray.
