Insulin and the insulin-like growth factors function in stem cell homeostasis across species with stem cell selfrenewal linked to cellular metabolism. Recent studies in the mammalian central nervous system support
essential roles for IGF/insulin signaling in neural stem cell self-renewal, neurogenesis and cognitive and
sensory function through distinct ligand-receptor interactions. IGF signaling is also known for its role in
myelination in the CNS. The PI3K/Akt/mTOR pathway is downstream of several receptors in the CNS including
the IGF/insulin receptors. mTOR signaling mediates a variety of processes in neural cells but has most recently
been investigated for its role in promoting oligodendrocyte differentiation and CNS myelination.

How cells control their size is a fundamental question in biology. Textbook hypotheses focus on mechanisms for sensing
size, time, or a combination of the two, based on classical correlation studies of the model bacterium Escherichia coli.
However, experimental evidence supporting either model was lacking due to the inability to assess the impact of changes
in size on single cells in real time. In this talk I will present all possible mechanisms in details. By analysing fluctutations in
physiological properties of division of hundreds of thousands of Gram-negative E. coli and Gram-positive B. subtilis cells
under a wide range of tightly controlled steady-state growth conditions, we are now in the position to validate different
theoretical models. The results demonstrate a strikingly simple mechanism whereby cells sense neither time nor space,
but add a constant volume each generation irrespective of their newborn sizes. This general mechanism quantitatively
explains all measurable aspects of growth and cell division at both population and single-cell levels, including the origin
and the hierarchy of variability in the underlying size-control mechanisms.

Mammary Stem Cells and Growth Control in the Breast

The overall goal of my laboratory is to understand how cells assemble into organs during development, and how the
reverse process occurs during cancer, when cells disassemble and metastasize to inappropriate locations. We use the
mammary gland as a model system to study two families of positional cues, called Slits and Netrins, which were
originally identified in the nervous system as axon guidance factors. We use both in vivo and in vitro techniques to
investigate how these cues control the development of the mammary gland, and how loss of these cues during tumor
progression contributes to breast cancer.

Dans la vague du Big Data, le mouvement vers les données ouvertes s’accélère. Les communautés scientifiques produisent depuis
longtemps de grandes quantités de données. La nouveauté est qu’elles devront à court terme les rendre accessibles, interopérables et
pérennes. * Les contextes scientifiques et techniques sont ceux du Linked Open Data et du Web Sémantique.
Les standards existent et les technologies connaissent un début de maturité.
Mais construire et pérenniser de grands ensembles de données exploitables, en particulier dans une perspective pluri- ou interdisciplinaire au delà d’une communauté restreinte reste un défi. *
Du point de vue institutionnel, la publication et la ré-utilisabilité des données de la recherche sont une exigence qui deviendra
rapidement incontournable. Le but de la journée est de présenter les questions scientifiques, les technologies disponibles et les
démarches institutionnelles associées à ces défis, à travers des exposés d’experts. Elle vise aussi à mettre en relation des actions
existantes qui participent au périmètre de l’Université Paris Saclay, et à explorer les voies qui peuvent mener à la création de services
mutualisés avancés, sous la forme de démonstrations et de posters, ainsi que d’une table ronde. Enfin, cette journée sera l’occasion de
présenter la plate-forme de partage de données que développe le Center for Data Science. Cette journée est organisée par le Center for
Data Science de l’Université Paris Saclay et l’Université Paris Sud

Abstract:
Infections by human pathogens such as HIV or HCV often vary in their course. For instance, in
absence of treatment some individuals infected by HIV-1 progress to AIDS rapidly, whereas others
remain asymptomatic for more than a decade. Similalry, some indiviuals manage to clear HCV
infections within a few weeks whereas others remain infected for life. Most of the research effort has
focused on identifying host factors that explain this variance from one infection to the next. This has
led to great successes such as the discovery of the role of IL-28 SNPs in clearing HCV infection or HLAB SNPs in controling HIV infection. However, these only expalin part of the variance. Recent results
show that a potentially strong fraction of the trait variation we observe could be explained by virus
factors. This largely rests on the fact that traits (such as set-point viral load or infection outcome)
seem to be "heritable" from one infection to the next. I will present these results along with three
explanatory hypotheses that could explain the somehow surprising "heritability" of HIV virulence.
Finally, I will present recent analyses of the ongoing Ebola virus epidemics in Sierra Leone to show
how phylogenetics can help us infer epidemiological parameters.

ABSTRACT: Enhancing immune responses with immune-modulatory monoclonal antibodies (mAbs) directed to inhibitory
immune-receptors is a promising modality in cancer therapy. Clinical efficacy has been demonstrated with antibodies
blocking inhibitory immune checkpoints such as CTLA-4 or PD-1/PD-L1. Treatment with ipilimumab, a fully human CTLA-4
specific mAb, showed durable clinical efficacy and improved overall survival in metastatic melanoma; its mechanism/s of
action, however, are only partially understood. We interrogated peripheral blood mononuclear cells (PBMCs) and
matched melanoma metastases from 29 patients treated with ipilimumab. Our findings show, for the first time, that
ipilimumab leads to the depletion of regulatory T cells (Tregs) via an antibody-dependent-cellular-cytotoxicity (ADCC)
mechanism, selectively mediated by FcgRIIIA (CD16)-expressing, nonclassical monocytes (CD14+CD16++). Our findings
provide novel mechanistic insight into the clinical activity of ipilimumab, highlighting the contribution of host-dependent
factors into the final outcome of antibody-based immune-modulatory therapies and identify nonclassical monocytes as a
potential biomarker of response.

MORE ABOUT DR. ROMANO: Dr. Romano is a medical oncologist-research scientist specializing in melanoma, tumor
immunotherapies, and early-phase clinical trials. In 2006, upon completing her studies at the University La Sapienza and
at the National Cancer Institute of Rome - Italy, she joined the Memorial Sloan-Kettering Cancer Center, where she
completed her postdoctoral training in tumor immunology and a fellowship in medical oncology leading to a position as
Senior Research Associate of the Dept. of Tumor Immunology. In June 2011, Dr. Romano joined the University Hospital
and Ludwig Institute for Cancer Research of Lausanne as a junior faculty member, where she was appointed attending
physician and group leader of the Laboratory Tumor Immunobiology of the Dept. of Oncology. Her current research
interests lie in the direct interrogation of the human tumor microenvironment by multiple biological and bioinformatics
approaches, the goal being to elucidate immune-protective and immune-escape mechanisms, and develop and
implement new modalities to harness the immune system to treat cancer

Neuron-microglial interactions are increasingly recognized as being key for physiological and
pathological processes in the central nervous system. Microglia have been found to play a causal role
in neuropathic pain behaviours resulting from peripheral nerve injury, and a core neuron-microglianeuron signaling pathway has been elucidated. Within the dorsal horn, microglia suppress neuronal
inhibition by a cascade involving activation of microglial P2X4 receptors causing the release of brain
derived neurotrophic factor (BDNF). BDNF acts on trkB receptors which leads to a rise in intracellular
chloride concentration in dorsal horn nociceptive output neurons, transforming the response
properties of these neurons. In addition to suppressing inhibition, peripheral nerve injury causes
activity-dependent facilitation at dorsal horn glutamatergic synapses which enhances nociceptive
transmission. This enhancement is mediated by intracellular signaling networks involving
serine/threonine and tyrosine kinases within nociceptive transmission neurons. Key for this
enhancement is facilitation of NMDA receptor function by Src family tyrosine kinases. Recently we
have discovered that microglia-to-neuron signaling is not only critical for pain hypersensitivity after
peripheral nerve injury but also for the paradoxical hyperalgesic effect of morphine and other
opioids. We anticipate that by targeting microglia-neuron signaling pathways new therapeutic
strategies for chronic pain as well as its comorbid sequelae may be developed.
