INTRODUCTION
Rapid Decalcifier
Stability and Storage
Raid Decalcifier is stable for two years. The bottles
must be kept closed. The advised storage temperature
is 18 - 30°C. Products that are past their shelf-life must
be disposed of, according to local disposal guidelines.
Procedure
A. Use in routine processes:
1. Use approximately 20-times the volume of the
specimen.
2. Allow to decalcify and continue the process.
B. Use on Microwave techniques:
We normally don’t recommend this technique for
clinical settings. Heating decalcifiers in a microwave
oven has only one major advantage: speed. In any
case care should be taken, because heat can burn
and destroy tissue and by doing so it won’t enhance
the quality of the specimens you are working with.
When using the microwave technique we advise

Performance Characteristics
The major advantages of the Rapid Decalcifier are:
Fast-acting
Bone Marrow biopsies are decalcified in as little as
15-30 minutes. Needle biopsies will be completely
decalcified in one hour or less. Small bone specimens and calcified arteries will be completely
decalcified in 2-4 hours. Typical surgical specimens
are completed in 2-6 hours.
Decalcification times are approximate.
Easy to control
Speed of processing can be controlled simply by
dilution with water.
Tissue protection
Ingredients are added to protect cells from actions
of acids.
Retention of Histological detail
Nuclear detail is preserved regardless of the speed
of decalcification.
Superior staining
Routine sections retain excellent staining characteristics. Limited range of recommended times
eliminate worry of impaired staining.
Non-hazardous and safe
The Decalcifier is a non-toxic and non-hazardous
solution, which can be spoilt in sewer systems
according local environmental regulations.
Furthermore the Decalcifier is safe in use.
STAINS AND DYES
Composition
Rapid Decalcifier is water based and contains
Hydrochloric acid, EDTA salts and surface active
components.
CLEARING
Principle
Decalcifying is necessary, because a high level of
calcium disturbs the staining quality. JT Baker’s
Decalcifier is a rapid decalcifying solution that has the
ability to decalcify the tissue completely within a short
time limit. We recommend using 20-times the volume
of the specimen. Following decalcification, the
specimen is ready for processing without first having
to wash. Tissues decalcified in this solution cut with
ease and routine HE techniques produce excellent
stained sections. Our experience is that decalcifiers in
general even work better if they are chilled.
1. Fix tissue in 4 w/v% Buffered Formalin. The
volume of fixative solution should be 10-times the
tissue volume.
2. Rinse fixed tissue in running water for at least 3
minutes. Immerse tissue in a volume of decalcifier equal to at least 10-times the tissue volume.
Microwave at 50% power until a temperature of
37°C is reached. Monitor this temperature closely.
Temperatures above 45°C seem to adversely
affect the tissue. For dense bone samples (teeth,
femoral heads, cortical bone), tissue should be
checked every 1⁄2 hour until an own time line is
developed.
For less dense bone (bone marrow biopsies)
tissue should be checked every 60 seconds until
an own time line is developed.

Intended Use
Rapid Decalcifier is intended to be used for decalcifying tissue sections such as bone marrow and bone
containing tissue sections.
