Charity Navigator Long Island Business News The Lustgarten Foundation has been recognized for the seventh consecutive year with the highest rating as a four star charity by Charity Navigator, the country’s premier independent charity evaluator – a designation that only 2 percent of the charities rated by this prestigious organization have received. Charity Navigator ratings demonstrate to donors how efficiently they believe a charity will use its support, and to what extent the charity is growing its programs and services over time. By adopting a focused research strategy, we have concentrated our efforts on working with world-renowned cancer research experts who have applied their knowledge and expertise to charting a new course for pancreatic cancer – a path that will lead to increasing survival rates and finding a cure. We are making substantial progress, in spite of limited government funding for pancreatic cancer research, and we are making it count for the patients who need our help now. In fact, 2015 marked a turning point for The Lustgarten Foundation, as we are on the cusp of exciting scientific breakthroughs from our Lustgarten-funded researchers in areas including immunotherapy and personalized medicine that could alter the course of pancreatic cancer and provide a renewed sense of hope and promise for pancreatic cancer patients and their loved ones. On the pages that follow, we highlight the noteworthy research we have funded with your support. We spotlight the groundbreaking studies being led by our Director of Research and Distinguished Scholar, Dr. David Tuveson, at The Lustgarten Foundation Pancreatic Cancer Research Laboratory at Cold Spring Harbor Laboratory. Dr. Tuveson and his team are conducting multiple studies with organoids, a reproduction of a human pancreas tumor, which will open new avenues for personalized medicine. Multiple drugs can be tested on a patient’s organoid to see which therapy shows the best response for the patient. This is the beginning of a new way of customizing treatment, offering hope to patients and dramatically improving patient care and prognosis. We also report on the ongoing research conducted by our other Distinguished Scholar award recipients, Dr. Bert Vogelstein, Johns Hopkins University School of Medicine, Dr. Ronald Evans, Salk Institute for Biological Studies, and Dr. Douglas Fearon, Cold Spring Harbor Laboratory and Weill Cornell Medical College. These accomplished researchers and our Research Investigators are conducting studies on early detection, immunotherapy, and potential new treatments that will impact outcomes. Additionally, we highlight our ongoing collaboration with Stand Up To Cancer, which now extends to four pancreatic cancer research teams that are tackling the disease using novel approaches. We want to take this opportunity to express our heartfelt gratitude to our many friends – our generous supporters, volunteers, walk leaders, event sponsors, Board of Directors, and Scientific Advisory Board. As a result of your dedication, we once again reached an all-time fundraising high of more than $20 million in 2015, and we held more than 300 community events and over 30 Pancreatic Cancer Research Walks nationwide. Vizza, Ph.D. Kerri Kaplan Chairman President & Chief Executive Officer Executive Director & Chief Operating Officer A Tribute To Cablevision A Vision Fulfilled In 1998, when a young 51-year-old Marc Lustgarten, Cablevision Vice Chairman, was diagnosed with pancreatic cancer, Charles F. Dolan and James L. Dolan, along with Marc, had a vision for a foundation dedicated to curing this difficult disease through research. Our mission remains steadfast — to advance research related to the diagnosis, treatment, cure and prevention of pancreatic cancer while raising public awareness and providing support for patients and their families. At the time The Lustgarten Foundation was established, pancreatic cancer was indeed an “orphan” disease, with less than $16 million, or 0.5 percent of the total National Cancer Institute’s budget supporting less than 15 researchers nationally. The founding fathers provided all the initial financial support to launch The Lustgarten Foundation. In 2008, in response to the leadership of Chairman Charles Dolan and Chief Executive James Dolan, Cablevision committed to pay all of The Lustgarten Foundation’s administrative expenses. This enabled the Foundation to publicly state that, “100% of every dollar donated goes directly to pancreatic cancer research” — a most powerful message. As a result, the impact on fundraising and research expenditures has been dramatic, with fundraising increasing threefold from $6.7 million to nearly $20.5 million in the period from the start of the sponsorship through 2015. Research expenditures increased fivefold from $19 million before Cablevision’s support to a total of $118 million through 2015, making The Lustgarten Foundation the nation’s largest private funder of pancreatic cancer research. In addition to this financial support, Cablevision created and supported our curePC campaign, which significantly raised awareness, provided patient information and helped increase fundraising. As a result, The Lustgarten Foundation’s impact on research has been substantial, supporting many major pancreatic cancer research milestones. Significant initiatives we have undertaken include: conducting research and clinical trials that led to the first new combination therapy in eight years; sequencing the genome; developing a personalized medicine model; developing an early detection blood test; harnessing the body’s own immune system to fight the disease; and developing a deeper understanding of the molecular biology underlying pancreatic cancer. ANNUAL REPORT • 3 Research In 2015, The Lustgarten Foundation initiated its OPT (Organoids for Personalized Therapy) trial, the first of its kind using human pancreatic cancer organoids to impact clinical care. This initiative is led by Dr. Tuveson and Project Manager, Herve Tiriac, Ph.D., at The Lustgarten Foundation Pancreatic Cancer Research Laboratory at Cold Spring Harbor Laboratory (CSHL). In 2014, Dr. Tuveson’s laboratory published findings on the first organoid model for pancreatic cancer in the prestigious journal Cell. Now, the team aims to apply this model to the clinic. David Tuveson, M.D., Ph.D., Director of The Lustgarten Foundation Pancreatic Cancer Research Laboratory at Cold Spring Harbor Laboratory, Professor and Deputy Director of the Cancer Center at Cold Spring Harbor Laboratory The first in a series of organoid-based projects, the primary goal of the OPT study is to demonstrate the effectiveness of the organoids as a precision medicine platform for pancreatic 4 • THE LUSTGARTEN FOUNDATION The team will then conduct a retrospective trial, in which they will compare the organoid’s responsiveness to the therapies to the patient’s actual clinical experience. In that way, the accuracy of the organoid as a predictor of drug response will be demonstrated. The next phase of the study will be a prospective trial in which the patient’s organoid will be tested prior to treatment to select what could be the most effective treatment for that patient. With a limited treatment window following diagnosis, there is an urgent need to correctly identify effective therapeutic options with the greatest likelihood to help the patient, making the development of the organoid critical. “The organoid represents a significant step forward for the future of cancer treatment,” said Dr. Tuveson. “We are confident that the OPT trial is just the beginning of a new way of customizing treatment at the time of diagnosis, offering hope to patients and dramatically improving the way that pancreatic cancer is managed.” Dr. Tuveson and his team have now made the organoid technology available to the entire pancreatic cancer research community through an educational course offered at Cold Spring Harbor Laboratory. Nearly 30 researchers have taken this class to date to learn how to create an organoid from a tumor in order to develop a personalized approach to treating patients. Our ultimate goal, therefore, is to make the first adaptable personalized medicine approach to treating pancreatic cancer, in the hopes of increasing survival rates substantially. To accomplish this goal, CSHL researchers will first collect 100 tumor tissues, from both patients who have undergone surgery and from the biopsies of metastatic patients, from several approved clinical sites throughout the country. The organoids will undergo a comprehensive evaluation, including genetic sequencing and therapeutic screening. Based on this evaluation, researchers will be able to characterize the patient’s organoid by its mutations and its responsiveness to over 120 FDA-approved chemotherapeutic and targeted therapies. Joseph Kellard Presently, it is difficult to determine how a patient will respond to a particular treatment, and therefore, the patient becomes the subject for testing. In the laboratory, the organoid becomes the subject for testing. The long-term goals of the organoid research project are to provide new, focused treatment options for patients based on their tumor’s genetic mutations. In this research project, the patient’s tumor organoid is genetically sequenced, and its genetic makeup is characterized. It is then “treated” with many compounds and combinations of compounds to determine which are the most effective in killing the cancer cells. This process — sequencing and screening drugs — is the essence of personalized medicine, where the precise treatment for each individual patient is selected based on the genetic makeup of the individual tumor. To date, a handful of patients have volunteered to have organoids developed for research purposes, and the preliminary results have been encouraging. In one case, a patient’s organoid did not respond to the current standard of care, and this result correlated to the patient’s actual response to those treatments. The organoid did respond well to a different therapy, and as a result, the patient was switched to that therapy. At the heart of our research program, David Tuveson, M.D., Ph.D., The Lustgarten Foundation’s Director of Research, and his team are focused on creating a personalized medicine platform using their recently developed “organoid,” which is a reproduction of a human tumor that offers hope to patients for dramatically improving their care and prognosis. An organoid can be reproduced indefinitely in the laboratory and then available drugs can be tested to determine which one(s) have the best response for a particular patient. We are confident that the OPT trial is just the beginning of a new way of customizing treatment at the time of diagnosis...” New Directions In Treatment Using Organoids For Personalized Medicine Joseph Kellard Interest in, and support for, pancreatic cancer research has grown substantially in recent years, as more scientists and clinicians are choosing to focus on this severely underfunded disease. Today, there are more than 1,000 researchers actively working to find a cure for pancreatic cancer, and there are nearly 750 pancreatic cancer clinical trials currently taking place across the country. The Lustgarten Foundation has significantly contributed to the growth of pancreatic cancer research by supporting more than 200 projects at more than 60 institutions worldwide. At the end of 2015, The Lustgarten Foundation’s commitment to pancreatic cancer research totaled more than $118 million since inception. In order to have the greatest impact on this disease, The Lustgarten Foundation has adopted a Focused Research Strategy — with emphasis on earlier detection, personalized therapy for individual patients and translational research to increase treatment options, which are highlighted in the research section that follows. ANNUAL REPORT • 5 The Lustgarten Foundation Expands Collaboration With Stand Up To Cancer (SU2C) The Lustgarten Foundation is pleased to recognize its long-term collaboration with Stand Up To Cancer (SU2C) which now extends to three pancreatic cancer Dream Teams and one pancreatic cancer research team engaging world-renowned scientists, with all projects involving clinical trials, leading to patient benefits in the three-year funding cycle. To date, The Lustgarten Foundation has committed more than $14.5 million to this collaboration for Stand Up To Cancer’s four pancreatic cancer research teams. We believe strongly that combining our resources and best practices with SU2C strengthens our research program and will move us more quickly towards significant progress. SU2C brings together the best and brightest minds to accelerate research so that new therapies can get to patients quickly and potentially save lives. The first pancreatic cancer Dream Team was one of the five inaugural 2009 SU2C Dream Teams funded from the first SU2C telecast in 2008. Led by Daniel D. Jaffee, M.D., Johns Hopkins University School of Medicine 6 • THE LUSTGARTEN FOUNDATION Additionally, the Dream Team developed a new imaging method to scan patients for tumors, based on the presence of glutamine, which provides the energy that the cancer cells need to grow and survive. The Dream Team’s research also demonstrated that early detection of recurrent pancreatic cancer was possible by testing blood samples for tumor DNA, called a “liquid biopsy.” Researchers determined that the blood tests were a more accurate way to learn if the disease had recurred following surgical removal of the tumor compared to standard-of-care imaging approaches. In fact, with the liquid biopsy, return of the disease was identified 6.5 months sooner than through imaging approaches. The SU2C-Lustgarten Foundation Pancreatic Cancer Convergence Dream Team: Transforming Pancreatic Cancer to a Treatable Disease, initiated in 2014 in conjunction with the Fox Family Cancer Research Funding Trust, aims to develop new therapies using patients’ own immune cells to treat their cancers through the five trials it is currently conducting. The team is also conducting studies to develop combination treatment approaches and biomarkers. It brings together leading individuals in the fields of immunotherapy, genetics, informatics, biostatistics, regulatory/clinical trials, cancer biology and pathology from nine institutions. Elizabeth M. Jaffee, M.D., Johns Hopkins University School of Medicine and Robert H. Vonderheide, M.D., D.Phil., University of Pennsylvania are, respectively, Leader and Co-leader of the project. In November 2015, the Stand Up To Cancer (SU2C) – Cancer Research UK – Lustgarten Foundation Dream Team: Reprogramming of Transcriptional Circuitry to Control Pancreatic Cancer, comprised of leading researchers from the United States and the United Kingdom, was announced at the AACR Molecular Targets meeting. This third Dream Team, led by Daniel D. Von Hoff, M.D., TGen, and co-led by Ronald M. Evans, Ph.D., Salk Institute for Biological Studies, and Gerard I. Evan, Ph.D., University of Cambridge, focuses on reprogramming pancreatic cancer cells so that they become less aggressive and more sensitive to drugs and anti-cancer immune responses. Through this research, the Dream Team’s goal is to improve the survival rate of pancreatic cancer patients, at least doubling the one-year survival in advanced pancreatic cancer patients from 35 percent to more than 70 percent, and to maintain remission in patients who have responded to treatment. A pancreatic cancer translational research team was formed under the direction of Jeffrey A. Drebin, M.D., Ph.D., University of Pennsylvania, to explore the effects of synthetic Vitamin D on the immune cells that enter the pancreatic cancer tumor. Dr. Drebin’s team will conduct a clinical trial of Gemcitabine, Abraxane and synthetic Vitamin D, followed by treatment with an immunotherapy drug that is currently FDA-approved for certain types of melanoma and lung cancer. Their research resulted in the discovery of combining Abraxane with Gemcitabine, which became the first new drug combination therapy approved for pancreatic cancer in eight years (since Tarceva was FDA-approved for use with Gemcitabine in 2005), as well as a standard of care for patients with metastatic cancer of the pancreas. Their research demonstrated that Abraxane combined with Gemcitabine increased one-year survival by 59 percent as compared to Gemcitabine alone. From left to right: Ronald M. Evan, Ph.D., University of Cambridge teams recently convened at the Stand Up To Cancer Annual Scientific Summit in California, where they had the opportunity to share ideas and best practices and provide feedback and suggestions for further research. This summit is particularly valuable in allowing researchers to share data, collaborate and achieve the best value in their research to potentially change the lives of thousands of patients and their families. A Look Ahead Our pancreatic cancer Dream Teams have already made significant strides in our understanding and treatment of the disease. Von Hoff, T Gen ANNUAL REPORT • 7 Distinguished Scholars Apply Novel Approaches To Early Detection And Treatment –B ert Vogelstein, M.D. Bert Vogelstein, M.D., Johns Hopkins University School of Medicine, is continuing to expand his research on a DNA-detecting blood test for pancreatic cancer to a larger, more diverse population. The goal of this research is to have a reliable blood test that can be administered at a doctor’s office during a routine appointment. Dr. Vogelstein is also studying the importance of surveillance in monitoring pancreatic cysts. In a successful study funded by The Lustgarten Foundation, Anne Marie Lennon, M.D., Ph.D., and her colleagues at Johns Hopkins University School of Medicine, led by Dr. Vogelstein, have identified a panel of molecular markers and clinical features that show promise for classifying pancreatic cysts and determining which cysts are likely to become malignant and require surgery, and which cysts are likely to remain benign and require monitoring only. This important finding was published in the November 2015 issue of the esteemed journal Gastroenterology. This more specific panel of markers will provide physicians with additional information to help them decide 8 • THE LUSTGARTEN FOUNDATION Focusing on more effective treatment for patients, Dr. Bert Vogelstein and his colleagues, Dr. Luis Diaz and Dr. Dung Le, recently published promising results in The New England Journal of Medicine using an immunotherapy drug called Pembrolizumab. Results demonstrated that cancer patients whose tumors had a unique mutation called “microsatellite instability” (MSI) responded to Pembrolizumab, even when other forms of therapy were not effective. There are potentially 2 percent or more pancreatic cancer patients who are MSI-positive and have the opportunity to potentially benefit from treatment with Pembrolizumab, which is not yet FDA approved for the treatment of MSI pancreatic cancer. As a result of the initial results with Pembrolizumab, The Lustgarten Foundation has launched a public awareness campaign to recruit patients and cover the costs to have their tumors tested for MSI so they have the opportunity to enroll in this promising clinical trial. Dr. Vogelstein also published an article in the American Association for Cancer Research’s journal Cancer Discovery highlighting the largest and most comprehensive genetic analysis of hereditary pancreatic cancer predisposition ever performed. Dr. Vogelstein and his team sequenced the genomes of 638 patients with familial pancreatic cancers and found that there are a number of different genes that explain familial pancreatic cancer, rather than just one gene, and that it is a highly genetically diverse disease. Ronald Evans, Ph.D., Salk Institute for Biological Studies, is closely investigating the relationship between pancreatic cancer cells and the cells in the stroma, the supportive tissue surrounding the tumor. Dr. Evans believes that the stromal cells send growth signals to the pancreatic cancer cells, and if these signals are blocked, the tumor will stop growing. Dr. Evans and his colleagues are testing how to do this, and are aiming to block the factors that promote the cancer. Thus far, they have discovered that by blocking a specific growth factor with a drug, a major cancer-causing gene is then blocked, and could prevent pancreatic tumor growth. Further research will be done to confirm if this will work in patients. In addition to this research, Dr. Evans continues to investigate the effectiveness of a derivative of Vitamin D in the clinical setting through his collaboration with Brian Wolpin, M.D., M.P.H., Dana-Farber Cancer Institute, and Eileen O’Reilly, M.D., Memorial Sloan Kettering Cancer Center. Drs. Wolpin and O’Reilly are performing standard biopsies on pancreatic cancer tumors. Dr. Evans then analyzes the Douglas Fearon, M.D., Cold Spring Harbor Laboratory and Weill Cornell Medical College, is conducting research to better understand the immune system’s role in pancreatic cancer. Pancreatic cancer cells can prevent a person’s immune system from attacking it by sending “stop” signals to the immune cells, which allows the cancer to evade cell death and continuously grow. Dr. Fearon’s research is investigating how immunotherapy drugs can turn these “stop” signals into “on” signals that can attack and kill the cancer. He currently has a clinical trial at Weill Cornell Medical College using immunotherapy drugs that sensitize pancreatic cancer cells to immune attack by boosting the tissue biopsy samples to establish a biomarker profile for patients whose tumors express the Vitamin D receptor. Tumors expressing this receptor are candidates for treatment with paricalcitol, a synthetic Vitamin D drug that targets and shuts down the “stromal cells” that surround the cancer cells and prevents chemotherapy from penetrating the tumor. Researchers were nominated by either the director of one of the 68 National Cancer Institute (NCI) designated cancer centers nationwide or by members of the Foundation’s Scientific Advisory Board (SAB). Award recipients each receive three-year grants of up to $1.5 million. We invite you to explore what each of our Research Investigators is studying: “There is absolutely hope in the fight against pancreatic cancer, and early detection is one of our most powerful tools for conquering the disease.” – Alison P. Klein, Ph.D., M.H.S., Johns Hopkins University School of Medicine Dr. Cantley’s research focuses on how a p53 mutation, the second most commonly mutated gene in pancreatic cancer, affects cell metabolism and cell signaling pathways that promote pancreatic cancer. Channing J. Der, Ph.D. University of North Carolina, Chapel Hill, NC Dr. Der’s study focuses on the proteins that KRas signals to. His research has revealed important findings about what makes a cancer cell sensitive or resistant when targeting the KRas pathway. This will help immensely as we aim to find a drug to target KRas, the mutation found in almost all pancreatic cancer patients. Stephen W. Fesik, Ph.D. Vanderbilt University Medical Center, Nashville, TN Dr. Fesik’s study aims to target other proteins that interact with the mutated KRas protein by using specially designed small molecule inhibitors that he created in hopes of indirectly killing KRas-expressing cancer cells. Charles S. Fuchs, M.D., M.P.H. and Mandar Muzumdar, M.D. Dana-Farber Cancer Institute and Massachusetts Institute of Technology, Boston, MA This research, being conducted simultaneously in humans and in mouse models, aims at better understanding how obesity and diabetes correlate to pancreatic cancer. Dr. Leach’s project uses a systematic approach to testing a very large number of different drug combinations in hopes of finding some that work best for certain groups of patients, based on specific biomarkers that would make patients appropriate candidates for particular drugs. Tony Hunter, Ph.D. Salk Institute for Biological Studies, La Jolla, CA Scott W. Lowe, Ph.D. Memorial Sloan Kettering Cancer Center, New York, NY Dr. Hunter’s study aims to more closely examine the relationship between cancer and stromal cells and identify pro-cancerous growth factors in hopes of targeting these factors to starve and kill the pancreatic cancer cells. Dr. Hunter’s research to date has demonstrated that when a specific cancerpromoting signal is blocked, mice with pancreatic cancer are living longer. Dr. Lowe’s research uses powerful genetic screening tools to identify genes that pancreatic cancer cells depend upon in hopes of more selectively and effectively killing cancer cells. In conjunction with Steven Leach, M.D., Dr. Lowe is using a new model for pancreatic cancer — the “speedy mice” — that develops pancreatic cancer more quickly than the conventional models, allowing for a significant number of drug combinations to be tested in a short time. Tyler Jacks, Ph.D. Massachusetts Institute of Technology, Cambridge, MA Dana-Farber Cancer Institute Dr. Jacks is using new-age genetic screening technology to discover more easily “druggable” gene products that mutant KRas-expressing cells are dependent upon to target, instead of KRas itself. He has discovered a way to reverse the cancerpromoting signals of KRas. Hidde L. Ploegh, Ph.D. Massachusetts Institute of Technology, Cambridge, MA Dr. Ploegh is developing a novel, non-invasive approach to imaging very small tumors, even smaller than what is detectable with the current standard of imaging. Stanford University School of Medicine Lewis C. Cantley, Ph.D. Weill Cornell Medical College, New York, NY Julien Sage, Ph.D. Stanford University School of Medicine, Stanford, CA Dr. Sage’s research targets a different class of proteins, known as KMTs, which play a role in pancreatic cancer progression. He has found a new cancer-causing protein that is a potential drug target, and he is now testing the effects of some therapies on this new target. Dr. Sage and his team are also focusing their research on combination therapies. If the therapies work together in a synergistic fashion, this could enhance the treatment response and reduce the chance of therapeutic resistance. Diane M. Simeone, M.D. University of Michigan Medical Center, Ann Arbor, MI Dr. Simeone’s study aims to sort and collect circulating tumor cells in a patient’s blood and then grow them in the laboratory for therapeutic testing and genetic sequencing. “We’ve learned a great deal about the molecular wiring of pancreas cancers, and we’re in a position now to take advantage of that knowledge with new treatment strategies.” – Tyler Jacks, Ph.D., Massachusetts Institute of Technology ANNUAL REPORT • 11 Cutting-Edge Research Influences Patient Outcomes With an overall survival rate of just 8 percent over five years, we don’t hear often enough about the courageous men and women who are beating these odds and giving a new face to the fight against pancreatic cancer. At this pivotal time in pancreatic cancer research, when we are on the cusp of potential scientific breakthroughs, we highlight below the inspiring stories of four pancreatic cancer patients and survivors who have benefited from Lustgarten-funded research that is having a profound impact on the detection and treatment of this disease. The Life-Changing Power of Immunotherapy Treatment In 2014, 52-year-old engineer David Blevins was diagnosed with pancreatic cancer that had metastasized to his liver. After a year of grueling chemotherapy treatment so debilitating that he was frequently unable to work, he threw his energy into finding a clinical trial. His research led him to Dr. Dung Le of Johns Hopkins University, and while he didn’t qualify for the initial trial he was hoping to participate in, Dr. Le convinced him to have his tumor tested for a rare MSI mutation (see page 8). On the same day that he was told by one of the many leading medical centers that he visited that there was nothing left to offer him besides chemotherapy, he received the life-changing call from Dr. Le that his tumor tested positive for the mutation. He was the first patient enrolled in a clinical trial for an immunotherapy drug targeting MSI, sponsored by The Lustgarten Foundation. Now, after more than 30 treatments over 14 months, David is doing phenomenally well. He is working again, experiencing minimal, manageable side effects and is fully engaged in his active family life with his wife and four children. At his most recent The Life-Saving Impact of Early Detection In 1994, retired surgeon Thomas Collins joined the National Familial Pancreas Tumor Registry at Johns Hopkins University, spearheaded by Dr. Ralph Hruban, a Lustgarten-funded researcher who focuses on early detection and familial pancreatic cancer. Tom was deeply concerned after several of his relatives, including his mother, passed away from pancreatic cancer. As part of the Lustgarten-funded CAPS (Cancer of the Pancreas Screening) research study led by Dr. Marcia Canto, a colleague of Dr. Hruban’s at Johns Hopkins University, Tom started making the trip from his home in Maine to Baltimore for an annual endoscopic ultrasound screening. Dr. Canto told him that few people continued coming for a screening for so many years; doing so most likely saved his life. During his eleventh screening in 2013, the ultrasound revealed a lesion the size of a pencil eraser – the smallest pancreatic cancer tumor to date discovered at Johns Hopkins. Tom underwent surgery and then 18 rounds of chemotherapy infusions. 12 • THE LUSTGARTEN FOUNDATION scan, there was no evidence of any more liver metastases, and his pancreas tumor had shrunk by more than 68 percent. And, because an MSI trial site recently opened at Ohio State University, he no longer needs to travel from his home in Indiana twice every month to Johns Hopkins in Baltimore for his infusions. Success with the First Pancreatic Cancer Vaccine David Blevins and his wife Carol Photo courtesy of David Blevins Kathleen Dowell was only 50 years old when she was diagnosed with pancreatic cancer in 1997, after doctors initially mistook her symptoms for gallstones. At first she was in complete shock and disbelief, as very little was even known about pancreatic cancer, and then she feared she would never live to see her newborn grandchild grow up. After undergoing surgery and chemotherapy, Kathleen was approached by Dr. Elizabeth Jaffee’s team at Johns Hopkins University to participate in a small study testing the first adjuvant pancreatic cancer vaccine (GVAX). She recalls her surgeon Dr. Cameron saying to her, “Mrs. Dowell, you could be the one to survive,” and those words were the encouragement she needed to try the vaccine. After just two GVAX doses, she became ineligible to continue due to complications from her prior chemotherapy treatment. However, she had the highest immune response of anyone in the study and now, 18 years later, she is cancer-free and has watched all four of her grandchildren grow up. The Lustgarten Foundation has funded Dr. Jaffee’s ongoing research on the GVAX vaccine. Dr. Ralph Hruban with Tom Collins Photo courtesy of Dr. Hruban In 2015, Jim Foley, a 71-year-old retired engineer and 15-year prostate cancer survivor from Westchester County, New York, was undergoing genetic testing to determine if he had the BRCA mutation, which two of his daughters and other family members have, when a routine CT scan for his long-term prostate cancer showed that he had a pulmonary embolism and metastatic pancreatic cancer. His entire world was turned upside down, as he learned that the embolism was caused by Stage IV pancreatic cancer that had metastasized to the liver. He was immediately referred to Dr. Eileen O’Reilly at Memorial Sloan Kettering Cancer Center, where he was also being treated for his prostate cancer. He learned that he tested positive for the BRCA mutation, which Dr. O’ Reilly said was actually beneficial because it gave her a roadmap for how to treat his tumor. She also said that BRCA patients often respond very well to standard of care chemotherapy. Now, a year since diagnosis, Jim shows no signs of slowing down. The tumor in his pancreas is not detectable and the lesions in his liver are dramatically reduced; in fact, his CA 19-9 and CEA tumor marker levels are now within the normal range. Jim experiences few side effects on the standard of care treatment regimen. Jim is busy spending his time with his wife, three daughters, and seven grandchildren. Jim also led Team Foley’s Fighters at the Foundation’s recent Westchester Pancreatic Cancer Research Walk, raising $3,300 for critical research. Several patients in a trial funded by The Lustgarten Foundation and led by Dr. Eileen O’Reilly at Memorial Sloan Kettering Cancer Center are having impressive results taking a PARP inhibitor in combination with cisplatin and gemcitabine. According to Dr. O’Reilly, the trial is likely to define a standard treatment approach for people with BRCA mutations. The Lustgarten Foundation is also funding the first comprehensive registry for people with a BRCA mutation, as well as those with a family history of BRCA mutations. This initiative is spearheaded by Dr. David Kelsen at Memorial Sloan Kettering Cancer Center. ANNUAL REPORT • 13 The Lustgarten Foundation and Cablevision Systems Corporation partnered in the curePC campaign, an award-winning public service campaign that raises awareness about pancreatic cancer and the Foundation’s efforts in the battle against this disease. 1 Team. 1 Mission. The curePC campaign’s series of online and print public service announcements (PSAs), “1 Team. 1 Mission.,” highlights how everyone’s contributions matter in the fight to beat pancreatic cancer. The series features the many team members – scientists, physicians, volunteers, caregivers and fundraisers – whose efforts help us make a difference. And on November 13, many Foundation supporters, including celebrities and members of the media, joined together for a special social media effort to mark World Pancreatic Cancer Day, reaching more than 12 million followers. On December 16th, Cablevision Systems Corporation, The Madison Square Garden Company, and AMC Networks hosted the 15th annual Holiday Rock & Roll Bash to benefit The Lustgarten Foundation. Held for the first time at the exclusive TAO Downtown in New York City, the Bash attracted over 1,000 people and raised more than $2 million for the second consecutive year, with 100 percent of every dollar raised going directly to support pancreatic cancer research. Since its inception in 2001, the Holiday Rock & Roll Bash has raised $21 million for pancreatic cancer research. On January 30th, more than 300 guests attended our 3rd annual New York City winter fundraiser, Party with a Purpose, held at the famous Marquee in the West Village. Guests enjoyed creative cuisine, danced to the sounds of DJ Matt Winters, participated in an auction with soughtafter prizes, and were entertained by a strolling magician and a dancing robot. Special thanks to our presenting sponsor Celgene and the dedicated event committee. Party with a Purpose garnered nearly $185,000, bringing the total raised from this event to more than $407,000. 16 • THE LUSTGARTEN FOUNDATION On February 6, the inaugural Race 4 A Cure took place at Grand Prix New York, an entertainment complex in Westchester County. Led by more than a dozen committee families from the surrounding community who have been impacted by pancreatic cancer, the event’s family-driven format brought together kids, teens and adults in the quest for a cure. With entertainment and music provided by Total Entertainment, indoor go kart racing, bowling, a bounce house and arcade, it was a fun-filled night for all. The fundraiser, emceed by Lisa LaRocca of News 12 Westchester, featured passed hors d’oeuvres from highly acclaimed local restaurants and a dinner and dessert buffet. Attendees also enjoyed a silent auction offering prizes including vacation packages, sports memorabilia, gift cards from top-rated restaurants, and show tickets and entertainment. The fundraiser attracted 400 people and raised more than $140,000 for The Lustgarten Foundation. We are proud to have worked with many dedicated supporters, patients and friends to increase awareness of pancreatic cancer and raise significant research funding through community events held across the country. We have highlighted some of our long-standing events below, as well as the people who have inspired them. Eddie Ivanoski Memorial Golf Tournament Village Cup Regatta The golf tournament honors the legacy of Edward “Eddie” Ivanoski, who served with the Marine Corps and as a Boston police officer and detective for 40 years. Eddie passed away from pancreatic cancer — one of five members of his family to die from the ravaging disease that also took the lives of his brother Leonard Ivanoski, two sisters Jean Morris and Irene White and nephew John Morris. The annual golf event, started in 2005, celebrates Eddie’s love for the sport and provides an opportunity for the Ivanoski family and their entire community to join together to raise money for a disease that has taken so much from them. To date, the event has raised nearly $229,000 for The Lustgarten Foundation’s research initiatives. The Village Cup Regatta was founded in 2010 by the Port Jefferson Yacht Club (formerly the Setauket Yacht Club) located on Long Island, New York as an annual event to raise funds for research related to the treatment and cure of pancreatic cancer. On September 12th, two teams — one comprised of Port Jefferson residents, and one made up of hospital staff — raced 30 boats amidst the cheers of hundreds of spectators. Actor/director Ralph Macchio and Maurice DuBois, who anchors the evening news for New York’s CBS affiliate and who grew up in Port Jefferson, served as the Village Cup Ambassadors. The Village Cup Regatta has raised $304,000 since inception, with funds shared equally between The Lustgarten Foundation and John T. Mather Memorial Hospital’s Palliative Care Program, located in Port Jefferson.
