The AgarClave serie is available in several models (10, 20, 40, 60, 2x20, 2x40, 2x60) basically differing in the useful volume of the sterilising well/wells. The control of the sterilising cycle is done by a microprocessor electronic card. The shaking speed of the culture medium is obtained using two pairs of metallic propellers adjustable in height. Their rotation is obtained thru the application of a rotating magnetic field on a magnetic rotor to which the propellers are linked. This magnetic linking avoids any possible leaking and is extremely easy to maintain. The shaking speed of the culture medium is largely adjustable. An alpha-numerical display gives to the operator information about the temperature and the status of the cycle. Up to 15 different sterilising programs can be set, with the possibility of inserting an after sterilisation constant temperature "cooking phase" for the addition of additives not bearing ordinary sterilising temperature. The building characteristics of this autoclave allow an easy and complete cleaning of the apparatus, which is very important considering the product to be handed. TKA Teknolabo A.S.S.I. Connect the unit to the water net after inserting a proper decalcification group thru nipple 1 in Pic. 3 and connect an outlet line to nipple 2 in Pic. 3, blocking the pipe with a ring. Pic. 3 Warning: there is hot water and/or steam exiting by nipple 2, use then appropriate pipes, interposing, when possible, a thermical trap. Insert the electrical plug, after having checked that the characteristics of the suit the ones on the label. It is important to make sure that the network is earthing provided and that the earthing is properly working. Do not use extensions or multiple shunts to feed this instrument. TKA Teknolabo A.S.S.I. In the inserting field a value is generally proposed: this value can be accepted or corrected as you please; the C button cancels one by one the present numbers starting from the one on the farthest right (last entered), the E button confirms the value shown on the display; Example: Presented value 1246, desired value 1346. Push C button, > 124 is shown Push C button, > 12 is shown Push C button, > 1 is shown Push 3 button, > 13 is shown Push 4 button, > 134 is shown Push 6 button, > 1346 is shown Confirm the value using the E button. Once you confirm it "Steril temp. > " is displayed, this refers to the sterilising temperature, then insert a value between 90 and 121 degrees. Once you confirm it "Steril temp. > " is displayed, this refers to the requested sterilising time, then insert a value between 1 and 60 minutes. Once you confirm it "Final temp. > (erogating temperature). " is displayed, this refers to the final temperature to keep If the set sterilising temperature is higher than 100 degrees, when confirming the "Adv. temp. > " writing is shown, referring to the temperature the additives have to be added at, insert a value between 30 and the previously setted sterilising temperature value (if this is not required, set as a value the same final temperature to be kept). When confirming "Adtv time > " is displayed, referring to the required "cooking" time, insert a value between 1 and 60 minutes. When confirming the "final temp. > " is displayed, referring to the final temperature to be kept (erogating temperature). Once this last parameter has been inserted the program is permanently kept in memory until a modificationt takes place, even if there is no power supply. TKA Teknolabo A.S.S.I. Via Quattro Strade 2 LACCHIARELLA (MI) +39 02 90033288 Fax +39 02 90033296 AgarClave User's Guide Page 17 USE: Before opening the well, make sure there is no pressure in the jacket (Pic. 5 ref. 2) and in the well (Pic. 6 ref. 1), otherwise release the pressure in the well thru the tap (Pic. 6 ref. 1) located on the cover of the well itself, and release the pressure in the jacket switching on the apparatus for at least 20 seconds. Warning: before performing the above operations make sure the autoclave has completely cooled off at the end of the previous cycle. Pic. 5 TKA Teknolabo A.S.S.I. The instructions herebelow refer to the models with screw knobs cover locking system. For the opening and closing of the models with cover central locking system please follow the instructions: -opening: rotate the central handwheel counter clockwise and rotate the locking bar in a convenient direction to access the working vessel. - closing: operate in the opposite direction. Please note that it is not necessary to apply an high rotating torque to obtain a proper closing as the gasket is double lip type. TKA Teknolabo A.S.S.I. Disconnect the nipple connecting the autoclave to the cover of the well (Pic. 7 ref.1-5) Pic. 7 Unscrew and remove the cover itself placing it on a surface, taking care not to damage the probeappendices or the shaker rod. Warning: for the models with removable baskets the well is subjected to the idrostatic pressure of the jacket, so when it is empty, if there is some water in the jacket (as it should be) it tends to float. Add the ingredients into the well (Pic. 8 ref. 1) Put the cover back in its place checking that the O-RING (Pic. 8 ref.2) is properly situated in its own place and block the cover using the 5 knobs (Pic. 7) taking care of screwing the knobs following the order:1-3-5-2-4. Picture 10 shows these 10 phases Pic. 11 The passage from a phase to the following one happens automatically, but when there are operations to be performed manualy, in this case the operator is alerted by a buzzer. It is possible to avoid manually some phases intervening on F1 button, but this opportunity has to be used only if it is compatible with a good performance of the cycle itself. Warning: In case of lack of electricity, the unit starts again automatically as the electricity comes back, therefore if a cycle has been launched, it will automatically restart. If you want to stop it, press the stop button that puts the autoclave in stand-by. In some of these cases it is advisable to stop the cycle, to restart it and pressing F1 bring it to a phase compatible with the gained temperatures. During the sterilising cycle the buzzer informs the operator when the well reaches a 96/97 degrees temperature, at this temperature make sure to close the exhausting tap on the cover of the well (Pic. 6 ref. 1). The operation has to be confirmed pressing the exhaust button. This operation is not necessary where the automatic exhaust option has been required. Afterwards the buzzer rings when the temperature for the addition of the thermolabile additives has been reached (cooking), if it has been previously set (see Programming). To confirm that the warning has been noticed press the exhaust button. At this point the reached temperature is indefinitely kept. Once the drawing of the product is ended, press the sop button. Warning: each time the operator press the stop the cycle is taken back to phase 0. If you want to start/restart from a different phase press F1 button to select the required phase. Information about the status of the autoclave are shown on the alpha-numerical display (Pic. 4 ref.2.) (i.e. jacket and well temperatures, electrovalves status and so on). Printer/Recorder of the thermical phase The preparator of the media is equipped with ALPHAPANEL printer/recorder which gives information regarding the temperature of the sterilizing well, the time of the beginning and ending of the cycle and any switch off of the equipment due to lack of power supply or voluntary action. Simply press the OPEN button (“key”) to start the recorder, once it is on the led is lit. Use the same button to switch it off. Press FEED to adjust the paper as desired. When the printing tape has reached the end and you want to re-use it, simply use your finger to rewind it (the tape is located in the internal right side). The recorder displays the day, month, year and hour. These parameters have been previously set by the manufacturer and they do not need to be modified. If a modification is required press OPEN before switching on the autoclave and release the button after pressing the main green switch. The ID unit serial number will appear on the display. Confirm this value using the “E” enter button and the year, month, day, hour, minute will appear on the display. If you need to replace the given values, use the “C” (cancel) button, insert the new values and confirm every single operation with “E” enter. The last thing on the display will be enter code>0. Confirm the 0 value with “E” to reach the operative stand-by phase. TKA Teknolabo A.S.S.I. The delay is shown in hour/minutes. To set this option follow this procedure: - press Select (instead of Start) insert the number of the program and press E insert the hour (0…99) insert the minutes (0…59) confirm with E, and the new time will appear for few seconds. The instrument stays on stand-by visualizing the clock, and at the preset time the cycle will start. Automatic exhaust This option makes completely automatic the preparation of the culture media. It is very important to make sure that the duct of the automatic exhaust is sterile. To achieve this make sure to install a proper hydrophobic bacterial filter, that can be sterilized in line (for ex. PALL C2PFRP1) on the exhaust side of the valve . To activate it, it is necessary to install this valve in its place using the supplied clamp pipe and link the connector of its excitation coil to the predisposed plug on the control head. Make sure the valve is properly installed with the arrow facing up. The valve itself can be sterilized and we strongly recommend this procedure if you operate in adverse conditions and /or with particularly sensitive media. In this case the sterilisation will be done with the bacterial filter installed on the ventilation pipeholder. The preparators equipped with this option can be used following the manual exhaust procedure. To do this simply replace the exhaust with the supplied clamp cap to close the door and follow the ordinary procedure. The system can automatically recognise whether the exhaust has been installed. Via Quattro Strade 2 LACCHIARELLA (MI) +39 02 90033288 Fax +39 02 90033296 AgarClave User's Guide Page 25 Acid pump (optional) The function of this pump is to introduce a certain amount of acid into the steam generator jacket at the end of every cycle to dissolve any possible lime deposit in the jacket itself. The amount of acid is defined by the manufacturer (TKA). The acid solution to be used is composed by citric acid 30%. WARNING: the control system cannot verify that during the phase of the introduction of the acid the acid itself is pumped into the jacked as there is no connection between the autoclave and the vessel with the diluted acid. For the instrument with removable well: - Open the well in the way described in chapter "Use" . Remove it in order to make the cleaning operation easier. Before putting it back check that the probes for the control of the water level in the jacket (Pic. 12 ref. 1 and 2) are in vertical position and that the O-RING (Pic. 12 ref. 3) is properly situated in its own place. For the instrument with fixed well introduce hot water (at least 50°) into the well itself, start the shaker for a few minutes and, at the same time, release completly the water. To better achive this use soft water and, periodially, acording to the toughness of the water, introduce citric acid in the jacket. Before the beginning of each cyle, as previously described, make sure that the space washer of the shating system is in good conditions (its thickness should not be less than 1 mm). If it is weared out replace it. TEKNOLABO A.S.S.I s.r.l. undertakes to freely repair or replace those parts that within waranty period Teknolabo itself should consider not efficient. The complete replacement of the apparatus and the extension of the warranty after an operation due to a failure are excluded. TEKNOLABO A.S.S.I. is not responsible for possible direct or undirect damages caused to people or things by failures of the apparatus or depending on forced suspension in its use. The warranty does not cover: aesthetic parts, damages caused by careless, bad or improper use or bad installation, mistaken connection to tension different from the one suitable to the apparatus, or different from the limitations fixed by CEI rules (+ -10% nominal value). Possible in laboratory-repairing is performed in our Technical Assistance Centre situated in Via Quattro Strade 2, - Lacchiarella - Milano unless different EXPLICIT indication or authorisation of the constructor. The warranty declines if the apparatus is tampered or repaired by nunauthorised personnel. The apparatus considered defective will have to be sent to us together with a copy of the certificate of warranty. The expenses and the risks of the travelling from and to our laboratory will be charged by the customer. In case of field repairing (to be performed only on constructor's not arguable decision), the customer has to pay the calling-right as a repayment for the travelling of the personnel. Validity of the warranty The waranty is valid only if the coupon has been fulfilled and has been sent to Teknolabo A.S.S.I. within 8 days from the date of the unit reception.
